Protonated Dipyridylamine Salts with Different Anions: Monomeric Tetraphenylborate as well as Bis(trifluormethylsulfonate), Dimeric Squarate and Polymeric Chloride DihydrateDi(pyrid-2-yl)amine is monoprotonated by acetic, squaric as well as hydrochloric acid and diprotonated by trifluoromethylsulfonic acid. To explore the anion and hydrogen-bridge dependency of its salts, crystals of the monomeric tetraphenylborate, the dimeric squarate as well as the bis(trifluoromethylsulfonate) and the polymeric chloride hydrate have been grown and their structures determined: Separated by bulky tetraphenylborate anions, pyrid-2-yl(2′-pyridinium) amine cation contains an intramolecular hydrogen bridge N⊕H…N and is slightly folded. The squarate salt dimer exhibits additional bridges, N—H…O⊖ and O—H…O. The severely twisted di(2-pyridinium)amine dication is connected to its trifluormethylsulfonate counter anions by altogether three H-bridges, two N⊕H…O and one N—H…O. The chloride dihydrate crystallizes in layers of 16-membered rings, formed from three Cl anions and five water molecules. Including these capabilities in a single instrument reduces construction costs and increases flexibility. AFM and LFM may be performed simultaneously; a simple reconfiguration (requiring removing the AFM/LFM cantilever holder and replacing with a STM tip) changes the instrument into a STM. Published 1995Staff ViewISSN: 1077-3118Source: AIP Digital ArchiveTopics: PhysicsNotes: This letter discusses the optical properties of Cr3+ ions in mixed borate crystals with the formula RX3(BO3)4, where R3+=Y3+ or Gd3+ and X3+=Al3+ or Sc3+. Measurements of the optical absorption spectra have been used to estimate the crystal field strength Dq and the Racah parameters B and C. These quantities confirm that in YAl3(BO3)4(YAB) and GdAl3(BO3)4(GAB)Cr3+ ions occupy strong crystal field sites 2E)〈E(4T2)], whereas in YSc3(BO3)4(YSB) and GdSc3(BO3)4(GSB) the Cr3+ ions occupy weak field sites 4T2)〈E(2E)]. The consequence is that the fluorescence spectrum of Cr3+:YAB and GAB reveals the sharp R lines at low temperature and R lines plus broadband at 300 K. Both Cr3+:YSB and GSB emit into the broadband 4T2→4A2 transition at all temperatures. The characteristics of the profiles within the mesophyll were determined by the optical properties of the leaf tissue and the spectral quality of the actinic light. When leaves were infiltrated with 10−4M DCMU [3-(3,4-dichlorophenyl)-1, 1-dimethyl-urea] or water, treatments that minimized light scattering, irradiation with 2000 μmol m−2 s−1 green light produced broad Gaussian-shaped fluorescence profiles that spanned most of the mesophyll. Profiles for chlorophyll fluorescence in the red (680 ± 16 nm) and far red (λ 〉 710 nm) were similar except that there was elevated red fluorescence near the adaxial leaf surface relative to far red fluorescence. Fluorescence profiles were narrower in non-infiltrated leaf samples where light scattering increased the light gradient. The fluorescence profile was broader when the leaf was irradiated on its adaxial versus abaxial surface due to the contrasting optical properties of the palisade and spongy mesophyll. Irradiation with blue, red and green monochromatic light produced profiles that peaked 50, 100 and 150 μm, respectively, beneath the irradiated surface. These results are consistent with previous measurements of the light gradient in spinach and they agree qualitatively with measurements of carbon fixation under monochromatic blue, red and green light. These results suggest that chlorophyll fluorescence profiles may be used to estimate the distribution of quanta that are absorbed within the leaf for photosynthesis.Type of Medium: Electronic ResourceURL: -

Previously, we have found that TPO induces a characteristic pattern of apoptosis during ex vivo expansion of human cord blood (CB) CD34+ cells and that the TPO-induced apoptotic cells belong to megakaryocyte (MK) lineage. In this study, we have examined the maturation of MK and platelet production in association with the TPO-induced apoptosis. CD34+ cells, purified from human CB, were expanded in serum-free conditions stimulated with TPO. Apoptosis was confirmed by terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate-biotin nick end labelling (TUNEL) assay and electron microscopy (EM). Simultaneous measurement of DNA content and immunophenotyping revealed that the cells with higher DNA content (〉8 N) constituted less than 5% of the CD41+ fractions until day 14, implying premature apoptosis of MKs before full polyploidization. Nevertheless, EM observation showed not only platelet territories but also newly produced platelets in which granules and microfilaments could be identified. Furthermore, flow cytometry demonstrated that the platelet fraction expressed P-selectin and an activation motif on GPIIb/IIIa recognized by monoclonal antibody PAC-1 upon stimulation with adenosine diphosphate (ADP). In addition, periodic acid-Schiff (PAS)-positive materials and nonspecific esterase activities could be demonstrated. Therefore, it is suggested that platelet production and the accompanying processes, rather than apoptosis only, be hastened during the ex vivo expansion of CB CD34+ cells when using TPO.Type of Medium: Electronic ResourceURL: -

Lysophosphatidylcholine (LPC), a phospholipid hydrolysis product produced by the activity of phospholipase A2, was detected in the lipids extracted from microsomes peroxidized by incubation with NADH and ADP-Fe3+. No detectable level of LPC was found in the microsomal lipids when NADH was withdrawn from the reaction mixture nor in the untreated microsomal lipids. LPC level in the lipids extracted from unheated microsomes was considerably higher than that from heat-treated microsomes when both were peroxidized by incubation with ascorbate and Fe3+. Phospholipase A2 was activated by either NADH-dependent or ascorbate-induced lipid peroxidation in vitro. The levels of lipid peroxidation in fish muscle were higher when fish were stored at higher frozen storage temperatures and so were the levels of LPC.Type of Medium: Electronic ResourceURL:
