Published 1996Staff ViewISSN: 1365-2133Source: Blackwell Publishing Journal Backfiles 1879-2005Topics: MedicineNotes: Summary We have investigated the effect of the synthetic phospholipid analogue, hexadecylphosphocholine (HePC), a member of a new class of membrane-affecting antiproliferative drugs, on adhesion to extracellular matrix components, reorganization of three-dimensional collagen lattices, and proliferative activity of human keratinocyte populations in vitro. Six transformed keratinocyte lines were compared with their normal counterparts from interfollicular epidermis.Adhesion to collagen types I and IV, laminin, and fibronectin was weakest in normal keratinocytes (binding of 3–10% of the cells), followed by HaCaT and HaCaT-II/3 (25-30% binding), and the squamous carcinoma lines and SV-40 transformed keratinocytes (35-50% binding). Treatment with non-toxic doses of 10–20 μmol/1 HePC led to a clear inhibition of the adhesive interactions by 50–75% in all populations. The impaired adhesion capacity was paralleled by a clear decrease of the ability of all keratinocyte populations to contract three-dimensional collagen lattices, an experimental mental model system for the reorganization of extracellular matrices. Histological examination revealed that these effects were due to a reduced cell number in the collagen lattices, a finding underscored by significant inhibition of proliferative activity of all keratinocyte populations by HePC. Furthermore, HePC led to marked changes of cellular morphology in the contracted gels. FACS analysis revealed that the impaired interaction with components of the extracellular matrix was not due to a specific downregulation of β1-integrins, the major cell-surface receptors for the respective matrix proteins.In conclusion, our results provide new insights into the potential action of HePC in a variety of skin disorders. Decreased proliferative activity and changes of cellular morphology, combined with impaired interactions with extracellular matrix proteins and a consecutive loss of matrix organization capacity, may cause suppression of different hyperproliferative skin diseases.Type of Medium: Electronic ResourceURL: -

In normal human skin, it is a characteristic marker for germ-cell phenotypic tissues. Although the gene encoding the MH 99 antigen has recently been cloned, and several histological and biochemical studies have been performed, the biological function of this interesting antigen still remains unknown.In the present study, we examined the synthesis of MH 99 in keratinocyte populations showing different in vitro differentiation capacity. Normal keratinocytes, spontaneously immortalized keratinocytes (cell line HaCaT), three SV-40-transformed keratinocyte lines (130, 425, and HaSV), and two squamous cell carcinoma lines (SCL-1 and SCL-2), were compared. Radioimmuno-precipitation revealed the highest levels of synthesis in cell populations with the least differentiation. This was paralleled by an increase of MH 99 synthesis in normal keratinocytes cultured in low concentrations of Ca2+ and by an increase of MH 99 synthesis during subculture of normal keratinocytes. Both phenomena were paralleled by an opposite behaviour of a differentiation marker. Molecular cross-linking and subsequent immunoprecipitation led to a decrease of the MH 99 signal, but an increase of a high molecular weight protein signal was seen. After cleavage of the crosslinker, the MH 99 signal reappeared, whereas the signal of the large protein remained unchanged. Thus, the MH 99 antigen may be associated with a high molecular weight protein on the cell surface, supporting the suggestion of a receptor-like function. Phosphorylation of the molecule could not be detected. Immunoelectron microscopy revealed homogeneous distribution on the cell surface, but cells of the same culture exhibited clear differences in their MH 99 expression.A concept for MH 99 regulation in normal and transformed human keratinocyte populations in vitro is proposed, showing that the synthesis of MH 99 is inversely correlated with cell differentiation. Distinct functional stages of dermal papilla cells (DPC) are involved in this process, thus suggesting that the dermal papilla is a highly specialized suborgan of the pilosebaceous unit. The aim of the present study was to investigate the functional properties of cultured DPC in various assays and to compare their functional properties with those of dermal fibroblasts (DFB). In monolayer cell cultures DPC showed an aggregative growth pattern, different to that of DFB, and lower proliferation rates, as compared to the controls. Adhesion assays performed using a 51[Cr]labelling method showed strong adhesion of both cell populations to collagen types I and IV, fibronectin and laminin, but DPC in vitro showed significantly higher adhesiveness to collagen type IV, a major component of the basement membrane of dermal papillae in vivo. The capacity of DPC to reorganize extracellular matrix components, as measured by gel contraction with three-dimensional collagen type I lattices, proved to be significantly lower than that of DFB and, moreover, DPC lysed the collagen lattices completely after 48 h in culture. The funtional differences between DPC and DFB were paralleled by higher surface expression and synthesis levels of the ‚1, ·1, and ·5 chains of integrin adhesion receptors in DPC, as detected by fluorescence-activated cell-sorter analysis and radioimmunoprecipitation. These findings provide evidence that DPC are a highly specialized cell population, which clearly differs from another mesenchymal cell type, DFB. After their isolation and cultivation in vitro, DPC still preserve functional properties related to important steps of cell-matrix interaction involved in the hair cycle.Type of Medium: Electronic ResourceURL: -

Publication Date: 2018-07-27Description: 'Der Beitrag vergleicht die Ergebnisse einer persönlichen Befragung einer Zufallsstichprobe, einer Online-Befragung zufällig ausgewählter Internetnutzer sowie einer offenen WWW-Umfrage zu politischen Einstellungen, die im Umfeld der Bundestagswahl 2002 durchgeführt wurden. Aus Unterschieden in der Rekrutierung der Respondenten und im Befragungsmodus werden Hypothesen abgeleitet und empirisch überprüft. Die Randverteilungen der Online-Befragungen, vor allem aber der offenen WWW-Umfrage, weichen deutlich von jenen in der mündlichen Befragung ab: Männer, junge, hochgebildete, politisch interessierte und informierte Menschen sind darin deutlich überrepräsentiert. Sieht man von Wissensfragen ab, lassen sich zwischen den drei Umfragen keine gravierenden Unterschiede im Messfehler nachweisen. In Bezug auf Reliabilitätsschätzungen treten nur geringe Differenzen zwischen den drei Erhebungen auf. Dagegen resultieren zu Korrelationen zwischen Variablen, die mit der Rekrutierungswahrscheinlichkeit in Online- und Offline-Erhebungen zusammenhängen, in den Online-Erhebungen, vor allem aber der WWW-Umfrage deutlich andere Befunde als in der mündlich befragten Zufallsstichprobe. Gleiches gilt für Korrelationen zwischen parteipolitischen Einstellungen und für die Erklärungskraft politischer Einstellungen in Bezug auf das Wahlverhalten. Online-Erhebungen, insbesondere aber offene Web-Umfragen sind somit weitgehend ungeeignet, valide Aussagen über Randverteilungen und Variablenzusammenhänge in der deutschen Gesamtbevölkerung zu treffen.' (Autorenreferat)'Online surveys - fast, cheap, but also valid? A comparison of two internet surveys with personal interviews on the German federal election 2002. This article analyses whether internet surveys on political attitudes allow for valid inferences on the electorate as a whole. For this purpose difference hypotheses on marginal distributions and correlations are proposed. They are tested empirically by comparing results of personally interviewed members of a probability sample, an online survey among members of an online panel and results of a survey among voluntary participants in an internet survey. As far as univariate statistics are concerned, the analysis reveals systematic biases in online surveys: Men, young, highly educated and politically interested persons participate in online surveys at a dis-proportionately high rate. Reliability estimates in online surveys do not diverge from results of personal interview data, but correlations among variables which are correlated with the selection probabilities in online surveys differ clearly from off-line survey results; additionally, online estimates of the correlations among political attitudes and the explanatory power of political attitudes on voting behaviour differ from offline results; and this is especially true for the www-survey. Hence, the results show that at the moment online surveys - and especially www-surveys - are of very limited value for the analysis of the electorate as a whole in Germany.' It was developed as a tool for inter- and transdisciplinary research on technology, sustainability and innovation. Its set of tools facilitates access to a scientific object from different perspectives, using different approaches. The methodology is characterized in particular by two factors: Heterogeneous factors of influence and their effectiveness in innovation processes are principally considered equivalent, existing in relation to one another and thus fitting together into constellations. Furthermore, the methodology facilitates a bottom-up approach, similar to the Grounded Theory Approach. According to this principle, constellations are described based on their singular units and their relationships to each other. The methodology is thus appropriate to the individual course of innovation processes. It enables the search for new patterns through the comparison of different innovation biographies and allows experimentation with strategic approaches as a special form of validation." Die Legenden fanden sich in Interviews, die ich mit Frauen führte, deren Männer das Dorf wg. Arbeitsmöglichkeiten in größeren Städten oder im Ausland verlassen hatten. Für die Analyse nutzte ich Immersion und offenes Kodieren (Marshall und Rossman 2006), wobei mich die individuellen Erzählungen und die Sinngebungsprozesse aufseiten der Frauen interessierten um nachzuvollziehen, in welcher Weise solche Legenden als signifikante Bestandteile des Alltagsleben wirksam sind (Gubrium und Holstein 2009). Die Erzählungen, die resultierten, entsprechen Goldsteins (2004) Definition einer Legende: Sie sind weder individuelle Geschichten, noch sind sie vollständig jenseits des Individuellen; sie werden im Duktus des 'Ob du es glaubst oder nicht' erzählt, und mit ihrer Erzählung sind nur minimale Risiken für die Erzählenden verbunden. Wie andere Legenden und Sagen dienen die hier Erzählten als Mittel der Stützung sozialer Ordnung in Zeiten sozialen Wandels. Und wie bereits in anderen Untersuchungen (z.B. Coontz 1999; Cooper, Linstroth und Chaitlin 2009; Greene 1991; Madriz 1997) deutlich, fokussieren sie insbesondere Frauen in sozialen Umbruchphasen. Die von mir interviewten Frauen berichteten auch über 'Unglücke' und 'Skandale', die sich dann ereigneten, wenn Frauen sich den Heiratsbräuchen oder der traditionellen Trennung von privaten und öffentlichen Räumen nicht unterwarfen." (Autorenreferat)"This article explores the folk legend as one articulation of the social control of women in rural Bangladesh. Stories and legends emerged when women were interviewed about the effects of men leaving the village for wage-based jobs in cities and abroad. Interviews were analyzed via immersion, theme generation, and open coding (Marshall und Rossman, 2006) with a focus on women's own narratives and meaning-making, which allowed for these stories to be understood as significant components of people's everyday realities (Gubrium und Holstein, 2009). These stories are consistent with Goldstein's (2004) definition of a legend: they are not personal stories yet they are not completely impersonal (friend-of-a-friend subject), they take on a believe-it-or-not tone, and telling them presents minimal risk to the narrator. Like other folk legends, these stories act as a means to reinforce the social order during a time of social change. As other research has shown (Coontz, 1999; Cooper, Linstroth und Chaitlin, 2009; Greene, 1991; Madriz, 1997), stories as social control mechanisms focus specifically and uniquely on women during times of social change. Respondents discuss 'accidents' and 'scandals' that occur when women do not adhere to marriage customs or the traditional dichotomy of public and domestic spheres." German-speaking scientists from all disciplines were invited to articulate their perspectives on principles and current problems in scientific publishing in the dialogical procedure. 697 participants addressed their opinion in two areas of consultation (a) Consultation area “evaluate principles”: the goal in this section was to find out whether there is a general consensus throughout academia of what constitutes a good publication system. For this purpose, principles of a good scientific publication system could be commented on and evaluated with positive or negative votes. (b) Consultation area “name problems”: this section aimed at obtaining the perspective of the participants on current challenges and problems of the publication system. Publication Date: 2018-08-31Description: Die niedersächsische Landtagswahl im Oktober 2017 fand im Schatten der vorhergehenden Bundestagswahl statt. Vor dem Hintergrund der zeitlichen Nähe beider Wahlgänge untersucht dieser Aufsatz die Entwicklung bundes- und landesbezogener Einstellungen im Wahlkampfverlauf sowie deren Einfluss auf das Wählerverhalten in Niedersachsen. Eine mehrwellige Wiederholungsbefragung von 1.346 niedersächsischen Wahlberechtigten, die im Rahmen der German Longitudinal Election Study (GLES) durchgeführt wurde, dient als Grundlage der Untersuchung. In quer- und längsschnittlichen Analysen wird gezeigt, dass eine bedeutende Minderheit der Befragten den Wahlentscheidungen auf der Landesebene ebenenspezifische Erwägungen zu Grunde legt. Landesbezogene Einstellungen waren einer von der Bundesebene weitgehend unabhängigen Wahlkampfdynamik unterworfen, die die Wahlabsichten des Elektorats beeinflusst zu haben scheinen. Publication Date: 2018-07-27Description: This paper addresses public opinion on common European policies in foreign affairs and defence. It proposes three models of support for common policies in these fields. Drawing on Eurobarometer data, the analysis shows that instrumental self-interest and territorial identities contribute considerably to explaining support for common foreign affairs and defence policies. Moreover, support for common policies is strongly driven by domain-specific evaluations of the European Union's performance. These findings suggest that popular support for common European policies in foreign affairs and defence has an experiential base. After a long period of stability, elections have dramatically altered their character - at first rather gradually, but during the past decade at an accelerated speed. Voters' decision-making has become much more volatile, rendering election outcomes less predictable. The long-term process of party system fragmentation that had already been going on for a while intensified sharply. A particularly conspicuous outcome of this period of turbulent electoral politics was the termination of Germany's exceptionality as one of the few European countries without a strong right-wing populist party. The chapter traces this development in detail. It identifies three distinctive phases of party system development: 1949 to 1976, 1980 to 2005, and 2009 to 2017. Published 2022Staff ViewPublication Date: 2022-11-17Description: This concluding chapter discusses changing German voters' behavior in the context of changing parties, campaigns, and media during the period of its hitherto most dramatically increased fluidity at the 2009, 2013, and 2017 federal elections. It summarizes the book’s findings on three questions: How did the turbulences that increasingly characterize German electoral politics come about? How did they in turn condition voters' decision-making? How were electoral attitudes and choices affected by situational factors that pertained to the specifics of particular elections? Discussing the consequences of these developments the chapter finds that the ideological and affective polarization of the party system has increased, leading to a dualistic structure that pits the right-wing populist AfD against all other parties. It also shows how the formation of governments under the German parliamentary system of governance gets increasingly difficult.
