Publication Date: 2018-07-27Description: Der Bericht gibt zunächst einen Überblick über Konzeption sowie organisatorische und wirtschaftliche Rahmenbedingungen des in Karlsruhe, Kassel, Schwerin und Stuttgart durchgeführten Modellprojekts, das im Folgenden evaluiert wird. Ziel des Projekts war es, mit niederschwelligen Konzepten zur beruflichen Förderung am modernisierten Verständnis der Hilfe zur Arbeit des Par. 72 BSHG anzusetzen und die arbeitsmarktpolitischen Angebote der verschiedenen Sozialleistungsträger so zu kombinieren, dass sich vor allem jüngere Frauen langfristig wieder ein wirtschaftlich selbständiges Leben aufbauen können. Das Projekt traf auf eine hohe Motivation bei den Teilnehmerinnen und kann als Erfolg gewertet werden. Die Teilnehmerinnen erwiesen sich als beschäftigungsfähig und reintegrierbar. Sie wiesen eine hohe Bildungsmotivation und ein hohes Interesse an betrieblicher Erprobung auf. Alle Angebote des Projekts wurden doppelt genutzt (berufliche Weiterentwicklung, Überwindung schwieriger Lebenslagen). Als unabdingbare Erfolgsvoraussetzung erwies sich die sozialpädagogische Begleitung. Published 2002Staff ViewISSN: 1600-0625Source: Blackwell Publishing Journal Backfiles 1879-2005Topics: MedicineNotes: Abstract: There have been contradictory reports on the shift in the T-cell cytokine expression pattern of peripheral blood mononuclear cells from patients with atopic dermatitis (AD); more specifically the interleukin (IL)-4 and interferon (IFN)-γ profiles. The aim of this study was to shed further light on this contradiction by measuring the intracellular cytokines IL-4 and IFN-γ by flow cytometry on unseparated whole blood to obtain results that, as accurately as possible, reflect the situation in circulating cells in vivo. The patient group including 64 patients with AD was compared with 18 nonatopic healthy adults. The results showed that the percentage of CD4+ T cells expressing IFN-γ was significantly decreased (P ≤ 0.001), as well as the percentage expressing IL-4 (P 〈 0.05) in AD patients compared with healthy controls. Furthermore, in supernatants from whole blood samples stimulated with phorbol 12-myristate 13-acetate and ionomycin, production of IFN-γ was significantly decreased, while IL-4 production remained unchanged in AD patients compared with healthy controls. We also investigated if there was a relationship between serum IgE level and Phadiatop®, a screening test for atopy, vs. the levels of IL-4 and IFN-γ , but found no correlation with either. However, there was a significant correlation between disease severity and the level of total IgE (r = 0.67, P 〈 0.05). The study exploited heterotrophic tobacco callus lines expressing a modified mammalian 6-phosphofructo-2-kinase/fructose 2,6-bisphosphatase that increased the fructose 2,6-bisphosphate content of the tissue. The phosphate status of two transgenic and one untransformed cell line was perturbed by incubation with 2-deoxyglucose, a phosphate-sequestering agent, and by growth of callus on phosphate-depleted media. 31P-NMR spectroscopy confirmed that both treatments decreased cellular levels of inorganic phosphate and phosphorylated metabolites. Despite large decreases in the amounts of phosphate esters, UDPglucose and adenylates in response to phosphate deficiency, the fructose 2,6-bisphosphate content of each line was unaffected by 2-deoxyglucose and increased during growth on phosphate-limited media. Short-term treatment of callus with 2-deoxyglucose had only minor effects on the carbohydrate status of each line, whereas long-term phosphate deficiency caused an increase in starch and a decrease in soluble sugar content in both transgenic and control lines. There were no consistent differences between the three callus lines in metabolism of [U-14C]glucose in response to incubation with 2-deoxyglucose. In contrast, there was a decrease in partitioning of label into glycolytic products (particularly organic acids) in untransformed callus during growth on phosphate-depleted medium. This decrease was greatly attenuated in the transgenic lines with increased fructose 2,6-bisphosphate content. This suggests that the conversion of hexose phosphates to triose phosphates is constrained under phosphate-deficient conditions, and that this restriction can be relieved by activation of pyrophosphate:fructose-6-phosphate 1-phosphotransferase. However, since the transgenic and control lines did not differ in the extent to which the carbohydrate content changed in response to growth on phosphate-depleted media, it is concluded that an increase in flux through pyrophosphate:fructose-6-phosphate 1-phosphotransferase is not a major component of the metabolic response of heterotrophic tobacco cells to phosphate deficiency.Type of Medium: Electronic ResourceURL: -
