Summary Returns of scheduled school entry examinations, from 838 primary school children in West Lambeth Health Authority, were analysed to assess the possible association of a child's home language background, age, sex, behaviour, and examiner with the results of the developmental examination. To allow for variation in social character-istics between participating schools, a ward index of deprivation based on the 1981 census was assigned to all the children in the schools of each ward. Variables representing development were most consist-ently associated with the child's behaviour during the examination and with the examiner. A child's home language background was associ-ated only with the auditory memory test. The child's ability to concen-trate during the test situation was closely related to his performance. With respect to the examiners, it is suggested that community health services need to consider schemes of ongoing in-service training and careful calibration of the tests.

Shock-induced luminescence from lithium niobate has been studied in the stress range 1.6–21.0 GPa. Both fast-framing photography and five-channel optical pyrometry were used to observe the luminescence. The framing photography showed that the emission pattern is heterogeneous for stresses just above the dynamic yield point. A further increase of the stress resulted in a pattern which was essentially homogeneous to within the experimental spatial resolution of about 30 μm. Narrowband filters and photomultiplier tubes were used in the optical pyrometry experiments. A broadband spectrum with a peak near 700 nm was observed. A plot of the energy dissipated by the shock versus shock stress correlates very well with a plot of the 700-nm intensity versus shock stress. The mechanism for light emission in lithium niobate appears to be closely related to the dynamic yielding process.

Forty-seven thymomas have been examined by DNA flow cytometry and results correlated with histology, stage, associated clinical features and survival. Twenty-five cases were DNA diploid, 14 were ancuploid and no results could be obtained for eight cases. The presence of ancuploidy correlated with more advanced (stage II and III) disease and the presence of myasthenia gravis and was more frequent in epithelial predominant thymomas. Tumour recurrence was more frequent in DNA aneuploid tumours, stage II/III disease and epithelial predominant neoplasms. Multifactorial analysis showed that DNA aneuploidy was predictive of tumour recurrence independent of the effects of stage and histology.

Immunohistochemistry using the PC10 antibody to proliferating cell nuclear antigen (PCNA) was applied to archival material from mucosa adjacent to gastric carcinoma (‘normal’, hyperplasia, complete and incomplete intestinal metaplasia and dysplasia) and non-cancer controls (normal and complete intestinal metaplasia). Overall, increased PCNA indices, with expansion and altered location of the proliferative zones, were observed in carcinoma fields and compared with controls (P≤0.001). These differences were particularly significant in ‘normal’ mucosa far from carcinoma as compared with normal in controls (P≤0.001). In carcinoma ‘fields’ distinct patterns of PCNA expression were noted in complete and incomplete intestinal metaplasia. Similarly, in dysplastic lesions high PCNA indices were present either throughout the gland or found predominantly in the upper compartment. We conclude that these differences in PCNA index and staining patterns might prove useful in monitoring the evolution of the disease in the follow-up of patients at risk of developing gastric cancer.

Abstract An examination of some characteristics of the blood plasma and urine of Trematomus bernacchii, an Antarctic fish, shows that at least in certain seasons the freezing points of these fluids are adjusted in response to environmental temperature. During October and November, freezing point of the urine was found to be about 0.7°C higher than that of the water. Adjustment of freezing point is seen to be intimately related to chloride concentration. At high temperatures magnesium concentration in the urine may be quite high; sodium replaces magnesium under cold stress. During cold stress, sodium and chloride move into the urine independently of each other.

Summary Unbiased estimators of genotype and allele frequencies and their respective variances are obtained for loci identified by mendelian segregation in haploid female gametophytes from individual trees. By a minimum sampling variance criterion, the allocation of experimental effort between the number of female gametophytes analysed per tree and the number of trees sampled per population is examined for a fixed total amount of experimental effort. For estimating heterozygosity, the optimum sampling design for many (generally most) cases is three female gametophytes per tree, but may be more than three depending upon the true genotype frequencies in the population. For estimating allele frequencies, the optimum sampling design is one female gametophyte per tree except in cases where a strong negative correlation exists between alleles within genotpyes. Guidelines are discussed for determining a suitable number of female gametophytes to be analysed per tree in order to estimate heterozygosity.

Summary Immunostaining to identify nuclear antigens expressed throughout the cell cycle provides a convenient way of assessing proliferating kinetics in tumours. We studied proliferation activity of gastric carcinomas by Ki-67 and PCNA immunostaining and the two methods were compared. The mode of tissue preparation differed, fresh frozen for Ki-67 and formalin-fixed paraffin-embedded for PCNA. Immunostaining with avidin-biotin was used in both. The labelling index (LI) and a semi-quantitative grading of cell proliferation were assessed in both markers. Significant correlation was shown between LI and grading with either Ki-67 and PCNA. However, no correlation was found between PCNA and Ki-67. This lack of relationship between the two markers may be attributed to a number of factors. 1. The most likely is the marked inter- and intra-tumour heterogeneity of gastric carcinomas reflected in high standard deviation values. 2. Preparation of tissue and small size sampling with Ki-67. 3. Long life of PCNA leading to detection of cells that have recently left the cell cycle. 4. One may be observing deregulated expression of DNA as seen in certain tumours. PCNA offers the advantage of being applicable to archival material.
