Exposure to these agents occurs through clothing such as undergarments and shoes, latex medical devices and latex and nitrile gloves. A simple, inexpensive screening method for total sulfur accelerator and a high performance liquid chromatographic speciation method were developed in the present study. These methods were applied to screen and quantify the sulfur accelerator content from 38 brands of ‘off-the-shelf’ latex and nitrile gloves obtained from commercial vendors. It was found that accelerator levels ranged from not detectable to 7.35 mg/g in the gloves analysed. Brands were found to contain single and multiple accelerator species within the glove. Powdered gloves had significantly higher accelerator levels than powder-free gloves from the same manufacturer; however, these chemical accelerators do not preferentially partition to the powder. The present analytical methodology is suitable for both manufacturing quality validation purposes, as well as for accelerator allergy research.Type of Medium: Electronic ResourceURL: -

Published 2002Staff ViewISSN: 1398-9995Source: Blackwell Publishing Journal Backfiles 1879-2005Topics: MedicineNotes: Background: Trimellitic anhydride (TMA)-induced occupational asthma is thought to be associated with its ability to acylate proteins and to induce production of TMA-specific immunoglobulin (Ig)E. Though the respiratory tract is considered to be a major exposure route leading to airway sensitization, the potential role of dermal exposure producing asthmatic sensitization is not known. The present study examines the ability of dry TMA powder to sensitize Brown Norway rats when applied, topically, to the skin. Methods: A patch of hair was carefully clipped with scissors on the rat's back. Dry TMA powder (0.3, 1.25, 5 and 20 mg) was administered on days 0, 7, 14 and 21, and the area occluded with surgical tape overnight after each application. Residual powder recovered from the occluded skin was analyzed by proton nuclear magnetic resonance and was still predominantly TMA. Circulating anti-TMA IgE and IgG were measured by ELISA.Results: TMA elicited dose-dependent production of specific IgE and IgG. Specific antibodies were detectable 2 weeks after the first TMA exposure and peaked between 3 and 4 weeks. Conclusion: The data suggest that topical skin exposure to dry TMA powder can induce allergic/immunological sensitization as demonstrated by the production of specific antibodies.Type of Medium: Electronic ResourceURL: -

ISSN: 1432-1793Source: Springer Online Journal Archives 1860-2000Topics: BiologyNotes: Abstract Female sand crabs (Emerita analoga) were sampled from 12 beaches along a 350 km stretch of Southern California coastline during the reproductive season in July, 1982. The percentage of mature females with egg masses and the condition of those egg masses were examined at each site. The proportion of mature females with egg masses varied only slightly between beaches. However, at sites within about 20 km of the San Onofre Nuclear Generating Station, a high percentage of females carried egg masses consisting of empty egg shells. A transplant experiment was conducted. Females with empty egg masses were taken from the 6.5 km N study site, where only 5% of the mature females carried normal egg masses, and transferred to running unfiltered seawater in Santa Barbara. After 2 wk, about a third of the transplanted females carried normal egg masses, while at the same time the proportion of mature females with normal egg masses remained at 5% on the beach of origin. Heated water discharged from the nuclear generating station is probably not the cause of the abnormal reproduction of sand crabs along the nearby coast. More likely causes are: (1) runoff of agricultural pesticides from a creek 3 km north of the nuclear generating plant; (2) release of metals from corroding cooling pipes, and/or (3) increased turbidity of the nearshore waters.Type of Medium: Electronic ResourceURL: -

Type of Medium: Electronic ResourceURL: -

Type of Medium: Electronic ResourceURL: -

The effect of paper dust collected at two different locations in a paper recycling plant (PD1 and PD2) on isolated nonsensitized guinea pig tracheal smooth muscle was studied in vitro. Dust extracts were prepared as a 1:10 w/v aqueous solution. Dose-related contractions of guinea pig tracheal rings were elicited with both PD1 and PD2. Pharmacologic studies were performed with atropine (10−6 M), indometacin (10−6 M), pyrilamine (10−6 M), LY171883 (10−5 M), nordihydroguaiaretic acid (10−5 M), and TMB8 (10−5 M). The possible role of endogenous neuropeptides in this constrictor process was studied by depleting neural mediators with capsaicin (5 × 10−6 M) before challenge with dust extracts. Constrictor effects were partially inhibited by a wide variety of the mediator blocking agents. The effects of both extracts were almost totally inhibited by the anticholinergic agent atropine, suggesting that a principal pathway mediating this response may involve the parasympathetic nervous system. The intracellular calcium-blocking agent TMB8 also induced a reduction of the contractile responses to PD1 and PD2 concsistent with the well established role of intracellular calcium in smooth muscle constriction. Pretreatment with capsaicin significantly increased the contractile activity of paper dust extracts but only at the higher doses of these extracts. This suggests that the effect of paper dust is not initiated by the release of mediators stored in sensory nerves but that the prerelease of these mediators may enhance the constrictor effects of these dusts. We suggest that paper dust extracts cause dose-related airway smooth muscle constriction possibly associated with the release of cholinergic as well as other mediators. The constrictor effect does not require tissue presensitization or the release of neuropeptides from sensory nerves.Type of Medium: Electronic ResourceURL:
