Published 1991Staff ViewISSN: 1471-4159Source: Blackwell Publishing Journal Backfiles 1879-2005Topics: MedicineNotes: Abstract: The neuron-specific synaptic vesicle-associated phosphoproteins synapsin I and synapsin II were shown to contain terminal 7V-acetylglucosamine (GlcNAc) residues as determined by specific labeling with bovine galactosyltrans-ferase and UDP-[3H]galactose. The β-elimination of galac-tosyltransferase radiolabeled synapsin I and subsequent analysis of released saccharide on high-voltage paper electrophoresis confirmed the presence of monosaccharidic GlcNAc moieties in O-linkage to the protein. Partial cleavage of synapsin I by collagenase, 2-nitro-5-thiocyanobenzoic acid, and Staphylococcus aureus V8 protease suggests that at least three glycosylation sites exist along the molecule. Taken together these data present the first evidence that a neuron-specific protein contains 0-glycosidically bound GlcNAc.Type of Medium: Electronic ResourceURL: -

ISSN: 1471-4159Source: Blackwell Publishing Journal Backfiles 1879-2005Topics: MedicineNotes: Abstract— The brain of a typical teleost fish, sea bass (Centrioprisles striatus), yielded subcellular fractions similar to those previously isolated from mammalian brain upon homogenization and differential and density gradient centrifuging. In particular, a well-defined synaptosomal fraction accounting for most of the bound acetylcholine of the tissue was obtained, although possibly because of the large amount of myelin in fish brain this and other fractions were not as homogeneous as the corresponding ones from guinea pig cerebral cortex. No difficulty was experienced in adapting the methods to the relatively small amounts of tissue (about 200 mg) obtainable from each fish. The isolation of synaptosomes in good yield indicates that teleost central presynaptic terminals have essentially the same physical properties as those of higher forms, permitting detachment and sealing under conditions of moderate liquid shear.Type of Medium: Electronic ResourceURL: -

Published 1975Staff ViewISSN: 1471-4159Source: Blackwell Publishing Journal Backfiles 1879-2005Topics: MedicineNotes: Abstract— An adenylate cyclase present in the caudate nucleus of rat brain, which is selectively stimulated by low concentrations of dopamine, and which is believed to mediate dopaminergic synaptic transmission, has been characterized with respect to several properties. The parameters studied included temperature, pH, ATP concentration, Mg/ATP ratio, and metal ion specificity. The effects of other compounds, including EGTA, NaF and several guanosine nucleotides, were also tested on the dopa-mine-sensitive adenylate cyclase. In addition, the subcellular distribution of the enzyme was studied. The highest specific activity was found in subcellular fractions enriched in nerve endings. A half-maximal increase in the activity of the enzyme in a subcellular fraction occurred in the presence of 4 × 10−6 M dopamine. Fluphenazine, a dopamine antagonist, competitively inhibited the activity of the enzyme in this fraction, with a calculated inhibition constant (Ki) of 8 × 10−9M.Type of Medium: Electronic ResourceURL: -
