    We have studied photoreflectance (PR) and reflectance spectra of undoped molecular-beam-epitaxy GaAs films on semi-insulating GaAs substrates at low temperature. In the PR spectra a sharp structure near the band-gap energy was observed which is sensitive to pump and probe light intensity. The origin of this structure was investigated by combining time-resolved and depth-resolved measurements. It is shown that the sharp PR structure arises from the superposition of a fast and a slow component of the modulated spectra. The slow PR signal originates from the epilayer–substrate interface. Both components are attributed to the modulation of exitonic transitions centered at slightly different energies. © 1995 American Institute of Physics.

    Below-gap structures in photoreflectance spectra of semi-insulating GaAs were observed originating from electroabsorption of light reflected from the back surface. The peak amplitude of these structures was strongly temperature dependent within the range (78–290) K. The different lineshape at low temperature in comparison to that at room temperature suggested different physical mechanisms. This conclusion was proven experimentally using a two phase lock-in technique by which a fast (X) and slow (Y) component of the spectra could be separated. A fit procedure provides the pure back surface reflection effect for both components. The X mode dominating at low temperatures is attributed to electromodulation of the field-broadened excitonic absorption tail whereas the Y mode originates from the Franz–Keldysh electroabsorption.

    The exact sine series method of Stein [Phys. Rev. A 43, 768 (1991)] has been extended to allow the calculation of critical Rayleigh numbers and wave numbers for both stationary and oscillatory convection states of rotating binary mixtures in Rayleigh–Bénard geometry. Checks on the accuracy of the method against previous calculations yield good agreement, provided these are multimode. Single-mode methods seriously underestimate Rc when the convection state is rotation-induced oscillatory. The method is used to obtain a hydrodynamic phase diagram showing a codimension-three point for a hypothetical mixture with Prandtl number 0.2 and Lewis number 0.0266. Threshold state calculations for a realistic 3He–4He mixture show that observation of a codimension-three point is impractical through the Prandtl number being too high.

    Gene mapping and marker-assisted selection in complex, polyploid genomes still relies strongly on restriction fragment length polymorphism (RFLP) analysis, as conversion of RFLP to polymerase chain reaction (PCR) markers can be very difficult. DNA extraction in amounts suitable for RFLP analysis represents the most time-consuming and labour-intensive step in molecular marker analysis of plant populations. In this paper, a new flexible method for plant DNA extraction is presented. It allows a high-throughput of samples in a short time without the need for freezing or lyophilizing the plant material. The method allows the isolation of genomic DNA with a yield of 100 μg for a minimal amount of 200 mg of leaf material. This is sufficient for work with large-genome plant species such as hexaploid wheat, where 20 μg of genomic DNA are required for a single RFLP analysis.
