    We report on our IR–IR double resonance experiment study of the excited stretching state of silane involving two or three vibrational quanta, and on the interpretation of our results by using the local mode model. The DR spectra were recorded by means of two separately tunable IR beams, one of which was used to pump selected lines inside the υ3 or 2υ3 IR bands, and the other (the probe) to observe transitions from the pumped vibration–rotation states. We characterized 7 out of the 14 vibrational states belonging to these manifolds for the first time, and the A1+F2 diad localized previously at 6362 cm−1 has been studied with much higher accuracy than before. Only the two highest states of the N=3 manifold are still unknown. The vibrational energies are very well reproduced by the local mode model ((approximately-equal-to)1 cm−1 rms deviation), and the harmonic interbond couplings have been determined more precisely than before. The rotational structures of the excited states are also well accounted for by Champion's theory of vibration–rotation coupling in Td, adapted to the description of stretching vibrations in terms of local modes. More high resolution (0.01 cm−1 or better) data are needed however to fully assess this point.

    By means of a pulsed slit jet and an infrared tunable diode laser spectrometer, the vibration–rotation absorption spectra of the complexes C2H2–H35Cl and C2H2–H37Cl have been observed for the first time in the 3.6 μm region of the ν1 band correlated with the HCl stretch. All the lines of the spectrum have been assigned for J=0 to 18 and Ka=0, 1, 2, 3. To determine the band origin and the rotational and centrifugal constants, the observed line frequencies have been fitted to those determined by the Watson Hamiltonian in the A reduction. A force constant model has been used to derive the binding energy De of the complex and the intermolecular stretching harmonic frequency from the experimental spectroscopic constants. The available experimental results concerning these complexes and other isotopic forms D35Cl and D37Cl were compared with ab initio calculations performed at the coupled-cluster single double triple [CCSD(T)] level of theory. The comparison turned out to be very good for all the properties considered (geometries, frequencies, energies). © 2000 American Institute of Physics.

    The different symptoms of resistance coded by the VF gene were studied on glasshouse-grown apple seedlings after inoculation with a conidial suspension of Venturia inaequalis. Scanning-electron microscopy and histological observations defined precisely three different classes of symptoms. Class 1 corresponds to a hypersensitive reaction. Class 2 is characterized by considerable tissue modifications but no sporulation whatsoever occurs; this reaction defines resistant hybrids. In class 3 the fungus remains active and can sometimes sporulate abundantly; this class was divided into two subgroups, one comprising all the hybrids of weak resistance, the other the hybrids of weak susceptibility.

    Abstract: We report an experimental study of aqueous foam imbibition in microgravity with strict mass conservation. The foam is in a Hele-Shaw cell. The bubble edge width ℓ is measured by image analysis. The penetration of the liquid in the foam, the foam imbibition, the foam inflation, and the rigidity loss are shown all to obey strict diffusion processes. The motion of bubbles needed for the foam inflation is a slow two-dimensional process with respect to the one-dimensional capillary rise of liquid. The foam is found to imbibes faster than it inflates.

    Abstract The external envelope glycoprotein (gp 160) of HIV-1 is a candidate for vaccines against AIDS. Most of the surface of the molecule is shielded by carbohydrate and the structures and locations of these glycans may be important in defining the immunogenicity of the viral coat. Here we report a sensitive mapping strategy for profiling and analysing the N-glycosylation of gp160, based on chemical release of glycans, fluorescent labelling and HPLC analysis. This approach has been validated in terms of establishing the reproducibility of all steps in the analytical procedure and on overall reproducibility on a run-to-run and day-to-day basis. The validated analysis technique was used to monitor the consistency of N-glycosylation of one rgp 160 vaccine candidate produced in bovine hamster kidney (BHK) cell culture. It was demonstrated that the variation in the glycan profiles of 6 different lots was not statistically significant.

    Abstract A clinical collaborative study was conducted to compare two new chromogenic agar media, Rambach agar and theSalmonella Detection and Identification Medium (SMID) (bioMérieux, France), with two conventional media, Salmonella-Shigella agar and Hektoen agar. Thirty-nineSalmonella strains involving 14 serotypes were isolated from 1,454 stool specimens. After enrichment in a selective broth, 100 % sensitivity was obtained with each medium. The SMID and Rambach agars are considerably more specific than the conventional media. Although SMID agar detects allSalmonella serotypes, it is not as specific as Rambach agar, which requires a complementary test (C8 esterase test) to detect all serotypes.

    This study deals with the structure and ultrastructure of the epithelial cells of the lizard (Lacerta vivipara Jacquin) epididymis as related to secretory activity. The epithelium contains only two types of cells, secretory cells and basal cells. The secretory cells undergo an annual cycle which has been divided into 10 stages. In its most active secretory state, epithelium forms 65.3% of the organ volume. The secretory cell is a tall columnar cell (from 55 ± 3.4 μm to 74.3 ± 2.4 μm height) with a basal nucleus and a supranuclear cytoplasm almost entirely occupied by numerous large secretory granules (5 to 7 μm in diameter). At the ultrastructural level, secretory cells contain rough endoplasmic reticulum (RER), Golgi complex, and secretory granules at various stages of synthesis before being discharged into the lumen. Each granule is membrane-limited and contains a spherical electron dense central core and a peripheral vacuole which varies in density. The secretory cell originates from small cubic cells (13.8 ± 0.7 μm) with few organelles (stage 1). The height of the cell increases gradually and free ribosomes appear first (stage 2), followed by scarce elements of RER (stage 3). The step preceding the secretion period (stage 4) is characterized by a conspicuous increase in volume of RER and Golgi complex. From stage 7 to stage 10, the cell undergoes a dramatic involution. After a transient hypertrophy of the RER, numerous autophagic vacuoles invade the cytoplasm. This degeneration can lead to a complete lysis of the cell and to its rebuilding after elimination of the greatest part of the cytoplasm. The volume of the epithelium falls to 15.6% of the total volume. With antibodies raised against the protein family which constitutes the main part of the secretion (L proteins of 19 kDa), it is shown by immunohistochemistry that these proteins are concentrated into secretory granules which are discharged into the lumen to finally bind to the heads of the spermatozoa.
