This singly measured biocontaminant concentration is related to adverse respiratory outcomes such as asthma, wheeze or others.Objective We analysed two repeated measurements of mite and cat allergens in mattress and living room floor dust as well as endotoxin concentrations in living room floor dust. The repeated samples were taken over a time period of about 6 years. We investigated the repeatability over time of their concentrations by determining correlation coefficients and computing within- and between-home variance components.Methods Our analysis was based on the population of a study on Indoor Factors and Genetics in Asthma (INGA) being carried out in 1995/96 and followed up in 2000/01. Complete data were available from 152 participants.Results The measured allergen concentrations were low and a considerable percentage of the values was below the limit of detection. The crude Spearman's rank correlation coefficients between the two measurements taken in 1995/96 and 2000/01 ranged from 0.32 to 0.61 for the dust mites allergens, from 0.21 to 0.44 for cat allergen and from 0.35 to 0.51 for endotoxin. Correlations were higher if measurements were performed on the same floor or the same mattress at both sampling time-points. The within-home variance for all measured biocontaminant concentrations was of about the same order as the between-home variance.Conclusion For studies like ours with low allergen and endotoxin concentrations, the repeatability of these concentrations over a time period as long as 6 years is low and a single measurement does not accurately reflect the true long-term exposure in the homes.Type of Medium: Electronic ResourceURL: -

Human THP-1 monocytes (106) were converted to macrophage phenotype by 500 ng/ml phorbol 12-myristate-13-acetate (PMA) and treated as follows: no stimulation or Escherichia coli LPS (10 µg/ml) alone or in combination with a physiologic dose of E2 (100 pg/ml) for 24 h in RPMI/10% FBS, washed extensively, then incubated for 24 h in serum-free media. Sujrenatant products were concentrated and incorporated into a 4% (w/v) methylcellulose gel. Separate gels were incorporated with the following: LPS (500 µg/animal) alone, high dose of E2 (10 ng/animal) alone, a combination of LPS+E2, or gel only (controls). Loaded or control gels were placed into a polylactic acid occlusive dome, inserted subcutaneously over the calvaria of mature ovariectomized ICR Swiss mice (8 mice×7 groups×2 times [5/14 days] = 112 animals), then calvaria were evaluated histologically. Macrophage stimulation with LPS alone, but not LPS in combination with E2, produced sujrenatants which upregulated osteoclast numbers in the suture area compared to gel controls at 5 days (p = 0.009). The addition of LPS directly to the local delivery gels significantly upregulated osteoclasts in endosteal surfaces compared to gel controls at 5 days (p = 0.024) and at 14 days (p = 0.025). The addition of E2 to LPS down-regulated resorption to a level not different from gel controls at 14 days. This in vivo model appears effective in studying inflammatory bone resorption, which may be inhibited by E2 directly or through its influence on secreted macrophage products.Type of Medium: Electronic ResourceURL: -

ISSN: 1432-1440Source: Springer Online Journal Archives 1860-2000Topics: MedicineNotes: Zusammenfassung Nach intravenöser Injektion von freiem Lactoflavin erfolgt in der Leber zunächst eine Anreicherung des Lactoflavins, die ihren Ausdruck in einer Zunahme der diffusen Grünfluorescenz findet. Sehr rasch darauf beginnt die Ausscheidung des Lactoflavins in den Gallencapillaren. Nach 2 Stunden ist der Ausscheidungsvorgang beendet. Auch in den Gallencapillaren ist das Lactoflavin lichtlabil, seine Fluorescenz wird in 18–30 Minuten durch die UV.-Strahlen ausgelöscht. Eine bestimmte Menge freien Lactoflavins wird aber weiter in den Leberzellen zurückgehalten. Sie findet sich als lichtlabile diffuse Grünfluorescenz der Leberzellen. Nach 6 Stunden ist das Lactoflavin auch in gebundener Form in der Leber sichtbar. In großem Maße sind die Kupfferschen Sternzellen mit einem gelbweißen, lichtlabilen Stoff beladen, der als an Eiweiß gebundenes Lactoflavin anzusehen ist. Mit Trypaflavin gibt diese komplexe Verbindung keine Rotfluorescenz. Von hier aus wird in die Leberzellen das Lactoflavin in seiner gebundenen Form abgegeben und neuerdings umgebaut. Die in den Leberzellen neu entstandene Lactoflavinverbindung gibt mit Trypaflavin eine hellrot leuchtende Fluorescenz, die bei UV.-Belichtung abblaßt.Type of Medium: Electronic ResourceURL: -

ISSN: 1432-0568Source: Springer Online Journal Archives 1860-2000Topics: MedicineNotes: Zusammenfassung Aus der Analyse der Architektur des Sinus sagittalis cranialis ergab sich ein Einblick in Anordnung und Wirken echter funktioneller Strukturen in der Dura mater cerebri. Ein Komplex von Faktoren konnte als einheitliches Gestaltungsprinzip in dem statisch-dynamischen Geschehen namhaft gemacht werden, der Dura und Sinus als zusammengehörige Einheit erfaßt. Das durale Bindegewebe erwies sich als plastische Masse, in der unterschiedliche Faktoren in einem vielfältigen Kräftespiel besondere Strukturen hervorrufen. Das Lebensalter stellt bestimmte Kombinationen solcher Faktoren zusammen, die in gesetzmäßigem Ablauf untereinander wechseln. Für das Einzelleben besteht ein weiter Spielraum in der Gestaltung möglicher Varianten, die jedoch niemals von einem übergeordneten Bauplan abweichen. Wesentliches Ziel der bindegewebigen Anordnung bleibt auch hier wie in anderen Organen die bestmögliche Anpassung des Gewebes an seine mechanische Beanspruchung. Nur in diesem Rahmen können die einzelnen morphologischen Befunde eine tiefere Deutung erfahren, die den Ausgangspunkt für den weiteren Weg in das physiologische und pathologische Geschehen bildet.Type of Medium: Electronic ResourceURL: -

In these the ferric ions are surrounded by weakly distorted octahedral complexes of water molecules. The relaxation process is characterized by several relaxation frequencies which replace the single flip frequency of earlier approaches. In contrast to the latter, they can be calculated from the microscopic properties of the sample. The only model parameter is the ligand field coupling constant D determining the relaxational behavior. A set of Mössbauer spectra, measured under various experimental conditions, is well described by a single value of the ligand field parameter D = 2.2gμB kG. Some characteristic features of the model become obvious in the spectra, e.g. the fading of certain absorption lines with increasing Fe3+ concentration.Type of Medium: Electronic ResourceURL: -

It has also become feasible to electrophoretically separate in two dimensions and to display DNA fragments derived from genomic digests. Through the appropriate choice of restriction enzymes, the functional component of the genome that encompasses CpG islands can be preferentially visualized in 2-D gels. The same computerized approach for the analysis of 2-D patterns can be applied to investigations at either the protein or DNA levels. Our group has utilized 2-D electrophoresis to investigate both protein and DNA changes in cancer. The emphasis to date has been on the identification of proteins, the abundance of which is related to specific biological features of the tumors analyzed and of DNA fragments encompassed in genomic amplifications, as the latter commonly contain growth-related genes. Findings derived from our analysis of neuroblastoma tumors and cell lines using 2-D approaches are reviewed. Data for four proteins observed in 2-D gels are presented because of our demonstrated association of these proteins with differentiation and proliferation properties of neuroblastoma. At the genomic level, the detection of amplifications using 2-D gels has necessitated an understanding of the variability displayed by multi-copy genomic fragments, which we have accomplished to a large part and which we present. An important benefit of 2-D approaches is the efficiency of scale and the ease with which abundant proteins or multicopy genomic fragments can be detected, identified and quantitatively analyzed.Additional Material: 3 Ill.Type of Medium: Electronic ResourceURL:
