    New grazing-exit electron-probe microanalysis (GE-EPMA) equipment is developed. In GE-EPMA, characteristic x rays are measured at the grazing-exit angle. X rays emitted from deep positions in the substrate are reduced under grazing-exit conditions; therefore, surface-sensitive analysis is possible with low background. In previous equipments, the sample holder was tilted to change the exit angle. In this new equipment, the energy-dispersive x-ray detector is moved to change the exit angle, and the analyzed position is stable even if the exit angle is changed. Therefore, this equipment is useful especially for particle analysis. The new GE-EPMA equipment is applied to Pd–Se–Te single-particle analysis. Although it was difficult to measure the Se Kα line at an exit angle of 45° due to the large Au Lβ radiation emitted from the Au substrate, Se Kα was measured without any Au signals at the grazing-exit angle near zero. © 2001 American Institute of Physics.

    We have developed a new analytical method, which we call the glancing-incidence and glancing-takeoff x-ray fluorescence (GIT-XRF) method. In this method, a fluorescent x ray is measured at various combinations of incident and takeoff angles. A nondestructive depth profiling is possible by using this GIT-XRF method, because the effective observation depth is changed by both the incident and takeoff angles. Here, we introduce the idea of depth profiling using the GIT-XRF method, and then we apply this method to an Au–Si interface reaction. © 1995 American Institute of Physics.

    To clarify the activation of peripheral blood T cells in Kawasaki disease (KD) patients, we investigated whether expression of lymphocyte function-associated antigen-1 (LFA-1, CD11a/CD18) and/or intercellular adhesion molecule-1 (ICAM-1, CDS4) on peripheral blood T cells increases during the acute stage. Expression of cellular adhesion molecules was measured using flow cytometry. There was a decrease in the percentage of CD3+ T cells in the bright LFA-1α and LFA-1β population and a concomitant increase in the dim population of LFA-1α and LFA-1β during the acute stage, in comparison with those of the convalescent stage. In addition, we observed no significant differences in ICAM-1 expression during the acute stage compared with that of the convalescent stage. In our view the present data, in conjunction with previous reports on T-cell function during acute KD, suggest that activated T cells are temporarily withdrawn from peripheral circulation during acute KD.

    Abstract— Usually, scatter in fracture toughness values is studied by assuming the employed steel plate to be macroscopically homogeneous in toughness, but it is probable that inhomogeneity contributes to scatter. This paper proposes a method which can discriminate scatter due to inhomogeneity from scatter measured in tests by using small size test pieces, cut out of previously fractured test specimens used for fracture toughness tests. By this method, the observed scatter of Kc(J) in a previous paper by some of the present authors was shown to be affected considerably by inhomogeneity, although the observed scatter was fairly well described by a Weibull distribution analysis.

    Background  Herpetic vesicles caused by herpes simplex virus and varicella zoster virus, and hydroa vacciniforme (HV) are characterized by umbilicated vesicule formation.Objectives  To understand the histogenesis of umbilicated vesicles in herpetic vesicles and HV, we demonstrated the presence of the virus-associated molecules in the lesions, and the pathogenic role of cytotoxic T-lymphocyte (CTL) immune responses.Methods  Phenotyping of infiltrating cells was carried out in biopsy specimens from herpes simplex, varicella, herpes zoster and HV, and compared with nonviral contact dermatitis. Viral antigens and Epstein–Barr virus-encoded small nuclear RNA (EBER) were detected by immunostaining and by in situ hybridization, respectively. Infiltrating CTLs expressing granzyme B and granulysin were determined by double immunostaining using confocal laser scanning microscopy.Results  In all herpetic vesicles, the corresponding viral antigens were observed in the cytopathic keratinocytes, and infiltration of lymphoid cells was present in the upper dermis and around the vessels. In all HV lesions studied, EBER+ T cells made up 5–10% of the dermal infiltrates and the dermal infiltrates contained almost no CD56 cells. CTLs expressing granzyme B and granulysin were present in both herpetic and HV lesions, in which they made up 10–30% of the total dermal infiltrates, whereas they comprised less than 5% of the infiltrates of biopsy specimens from nonviral contact dermatitis. Confocal laser microscopic examination demonstrated that both CD4+ and CD8+ T cells expressed granzyme B and granulysin.Conclusions  CD4+ and/or CD8+ CTLs reactive to the virus-infected cells might be responsible for the histogenesis of herpetic and HV lesions characterized by umbilicated vesicles.
