It has recently been shown that the molecular pharmacology of CGRP and ADM is determined by coexpression of one of three receptor activity-modifying proteins (RAMPs) with calcitonin receptor-like receptor (CRLR). Furthermore, RAMP proteins have also been shown to govern the pharmacology of the calcitonin receptor, which in association with RAMP1 or RAMP3, binds amylin with high affinity. In this study, we have cloned the rat RAMP family and characterized the pharmacology of rat CGRP and ADM receptors. Rat RAMP1, RAMP2 and RAMP3 shared 72%, 69% and 85% homology with their respective human homologues. As expected CRLR-RAMP1 coexpression conferred sensitivity to CGRP, whilst association of RAMP2 or RAMP3 with CRLR conferred high affinity ADM binding. Using specific oligonucleotides we have determined the expression of RAMP1, RAMP2 and RAMP3 mRNAs in the rat central nervous system by in situ hybridization. The localization of RAMP mRNAs was heterogeneous. RAMP1 mRNA was predominantly expressed in cortex, caudate putamen and olfactory tubercles; RAMP2 mRNA was most abundant in hypothalamus; and RAMP3 was restrictively expressed in thalamic nuclei. Interestingly, in specific brain areas only a single RAMP mRNA was often detected, suggesting mutual exclusivity in expression. These data allow predictions to be made of where each RAMP protein may heterodimerize with its partner G-protein-coupled receptor(s) at the cellular level and consequently advance current understanding of cellular sites of action of CGRP, ADM, amylin and CT. Furthermore, these localization data suggest that the RAMP family may associate and modify the behaviour of other, as yet unidentified neurotransmitter receptors.Type of Medium: Electronic ResourceURL: -

We investigated whether intraocular use of thymoxamine 0.02% reduced the number of endothelial cells. Patients and methods: After phacoemulsification of 59 eyes, either thymoxamine 0.02%, acetylcholine 1.0% or buffered saline solution was given intraocularly. With a contact specular microscope, corneal endothelial cell photographs were taken on the day before treatment and 3 days and 6 weeks after surgery. Results: There were no statistically significant differences between endothelial cell counts of eyes treated with thymoxamine (–7.2%), acetylcholine (–10.2%) or BSS (–9.4%). Conclusion: This study shows for the first time that thymoxamine, when given in the anterior chamber after phacoemulsification, does not cause a greater loss of endothelial cells than acetylcholine or buffered saline solution.Notes: Hintergrund: Das Miotikum Thymoxamin, ein α-1-Rezeptoren-Antagonist mit sympatholytischen Eigenschaften, wirkt bei intraokularer Applikation additiv zu Azetylcholin, so daß die Pupillenkonstriktion stärker ist als bei alleiniger Gabe einer der beiden Substanzen. Wir untersuchten die Auswirkung von intraokular appliziertem 0,02%igem Thymoxamin auf die Endothelzellzahl in einem randomisierten, prospektiven Doppelblindvergleich, um herauszufinden, ob bei Anwendung von Thymoxamin eine Endothelzellzahlreduktion eintritt. Patienten und Methode: Bei 59 Patienten wurde nach einer Phakoemulsifikation entweder Thymoxamin 0,02% oder Azetylcholin 1,0% oder gepufferte Elektrolytlösung (BSS) intraokular appliziert. Das zentrale Hornhautendothel wurde präoperativ, 3 – 5 Tage postoperativ sowie 6 Wochen postoperativ kontaktoptisch fotografiert. Die Endothelzellparameter wurden jeweils anhand von mindestens 50 Endothelzellen ermittelt. Ergebnisse und Schlußfolgerungen: Anhand der gewonnenen Daten konnte erstmals gezeigt werden, daß nach Phakoemulsifikation und Hinterkammerlinsenimplantation intraokular angewendetes Thymoxamin (–7,2%) nicht zu einer stärkeren Endothelzellreduktion führt als die Anwendung von 1%igem Azetylcholin (–10,2%) oder BSS (–9,4%).Type of Medium: Electronic ResourceURL: -

The data were compared with results found on NC 203 SiC. The temperatures used to conduct the tests ranged from 595–1049°C, and this information complemented data collected earlier. The main results are as follows: the modulus for RFSC specimens decreases with increasing temperature over a range of approximately 350–260 GPa from room temperature to 1049°C, the modulus for NC203 ranges from 450–415 GPa over the same temperature range, and damping of RFSCs is independent of strain amplitude and has little temperature dependence.Type of Medium: Electronic ResourceURL: -
