    Abstract Bispecific antibodies with specificity for CD3 and a tumor antigen can redirect cytolytic T cells to kill tumor targets, regardless of their natural specificity. To assess the clinical potential of bispecific antibodies for treatment of human cancers we have, in the present study, adapted a totally syngeneic mouse model to the targeting of mouse T cells against mouse tumors in immunocompetent mice. We show that gp52 of the mouse mammary tumor virus (MTV) can serve as a tumor-specific antigen for redirected cellular cytotoxicity. Chemically crosslinked and genetically engineered bispecific antibodies with specificities for gp52 and murine CD3 ε-chain induced activated mouse T cells to specifically lyse mouse mammary tumor cells from cultured lines and primary tumors from C3H-MTV+ mice. Retargeted T cells also blocked the growth of mammary tumors in vitro as well as their growth in syngeneic mice. These findings identify murine MTV-induced mammary adenocarcinomas as a solid-tumor, animal model for retargetin T cells with bispecific antibodies against syngeneic breast cancer.

    Resonant-optical-waveguide (ROW) arrays with a self-aligned-stripe (SAS) geometry have been realized for the first time. Resonance is achieved by holding the array element/interelement widths constant and varying the aluminum composition of the passive guide layer. External differential quantum efficiencies as high as 47% have been achieved from longitudinally uniform 20-element 1000 μm-long devices. Diffraction-limited-beam operation is obtained up to 13×threshold, and 2.1 W output power level from devices with Talbot-type spatial filters. The front-facet emitted coherent uniphase power is 1.6 W, with 1.15 W in the central lobe.

    Superconducting MgB2 thin films were prepared on Al2O3(0001) and MgO(100) substrates. Boron thin films were deposited by the electron-beam evaporation followed by postannealing process with magnesium. Proper postannealing conditions were investigated to grow good superconducting MgB2 thin films. The x-ray diffraction patterns showed randomly oriented growth of MgB2 phase in our thin films. The surface morphology was examined by scanning electron microscope and atomic force microscope. Critical current density (JC) measured by transport method was about 107 A/cm2 at 15 K, and superconducting transition temperature (TC) was ∼39 K in the MgB2 thin films on Al2O3. © 2001 American Institute of Physics.

    GaN epilayers grown by molecular beam epitaxy were photoelectrochemically (PEC) oxidized in an aqueous KOH solution. The oxidation effect was investigated by defect-related photoconductivity and photoluminescence. The PEC treated GaN show decreased extrinsic photoresponse and concentration of deep level states in comparison with the as-grown sample. The PEC process also results in enhanced donor-bound exciton photoluminescence at 3.47 eV and restrained 3.4 eV band. No strain is detected in the PEC oxidized GaN. The 3.4 eV band is related to structural defects instead of oxygen impurities. Rather, the defects can be passivated by the PEC oxidation. © 2001 American Institute of Physics.

    The Rheological behavior of mozzarella cheese filled with various proteins (whey protein, caseinate, egg white, soy protein isolate, gelatin) incorporated was determined by uniaxial compression at 10°C and the effect of temperature (10°C−60°C) by dynamic measurement. Mozzarella cheese with whey protein, caseinate, egg white, and soy protein isolate showed significant water retention during heating. Among the proteins, soy protein isolate induced the strongest gel network structure with mozzarella cheese. All proteins altered the viscoelastic properties of mozzarella cheese.

    Cheeses with whey pH at draw of 6.15 or 6.40 were stored at 4°for 50 days. Compression test at 10°(Instron) showed a decrease in compressive stress with storage. Failure points were near a true strain value of 0.5 for the cheeses at day 3. The failure region became less obvious by day 50. The hypothetical equilibrium stress (HES) in the stress-relaxation test was higher with a draw pH of 6.40 than 6.15, possibly indicating that higher calcium led to a stronger network structure. The HES decreased with storage, suggesting cleavage of structural crosslinks caused by the coagulant.

    Growth and aflatoxin production by injured spores (heat, 55°C for 15 min, γ-irradiation, 50 Krad) of Aspergillus parasiticus in the presence of sorbate (0.05 and 0.1%) and propionate (0.2 and 0.4%) were studied in YES broth at pH 4.5, 5.5 and 6.5, and 25°C for 21 days. Aflatoxin production was accelerated in the early stages of growth by γ-irradiation, but not heat. Growth and aflatoxin were delayed in cultures from low numbers of uninjured spores. Aflatoxin increased and was produced sooner by low numbers of uninjured spores in 0.05% sorbate. Both inhibitors slowed growth of injured spores more than non-injured. Inhibition of aflatoxin was dependent on the concentration of inhibitor, pH and injury.

    Summary Consider the solution of one-dimensional linear initial-boundary value problems by a finite element method of lines using a piecewiseP th -degree polynomial basis. A posteriori estimates of the discretization error are obtained as the solutions of either local parabolic or local elliptic finite element problems using piecewise polynomial corrections of degreep+1 that vanish at element ends. Error estimates computed in this manner are shown to converge in energy under mesh refinement to the exact finite element discretization error. Computational results indicate that the error estimates are robust over a wide range of mesh spacings and polynomial degrees and are, furthermore, applicable in situations that are not supported by the analysis.

    Summary The thyroid glands of transgenic mice (TM) expressing the genes for human (h) and bovine (b) growth hormone (GH) were studied. The percentages of larger follicles inhGH TM of either sex were significantly greater than in the corresponding normal littermates, and follicles ranging up to 350 μm in diameter were present in malehGH TM. In contrast, thyroid follicles were only slightly enlarged in malebGH TM, and were unchanged in femalebGH TM. The serum concentrations of T4 were significantly decreased in malebGH TM and not altered in the other groups. Serum concentrations of T3 were slightly, but significantly increased in femalehGH TM and femalebGH TM, but were unaffected in male TM of either type. Since the principal difference between these foreign GHs in rodents is the additional lactogenic activity of human GH, these results may indicate that the effects of prolactin can influence the development of the thyroid.

    Abstract A three-dimensional steady incompressible viscous flow through a plane cascade of turbine blades has been analyzed through a numerical method based on the Navier-Stokes equation. Particular attention is paid to the prediction of secondary flows occurring due to the endwall boundary layer and the blade geometry. A standard k-ε model is used for the modelling of Reynolds stress and boundary-fitted coordinates are adopted to represent the complex blade geometry accurately. Two differencing schemes are applied to the convective terms to investigate the effect of numerical diffusion. Experimental data obtained for the flows through the Langston cascade are selected for code validation. Computed results for the velocity vectors and static pressure distributions are in good agreement with presious measurements and provide validity of this numerical method. Three-dimensional viscous flow phenomena and the distribution of total pressure loss caused by secondary flows are also reasonably well predicted.
