Since tritium is the principle radionucleid released from thermonuclear tests its effects on the most sensitive phases of mammalian life, the fetus, seemed worth investigation. Our study was undertaken to approximate a realistic situation, the consequenses to the development of fetal hemopoiesis after maternal ingestion of HTO during pregnancy. Doses of 5800 μCi/day resulted in a significant impairment of the development of fetal hemopoiesis in the newborn animals, during the entire observation period of 3 weeks after birth. Litter size and post-natal death rate was not altered. Smaller doses than 5800 μCi/day had no significant effects on the hemopoiesis of the offspring except a decrease of lymphocytes demonstrable after 2800 μCi/day HTO. A comparison with dates obtained after3H-thymidine (3H-TdR) infusion reveals that tenfold higher doses of HTO are necessary to demonstrate similar effects in the offspring. The mechanisms for this phenomenon are discussed, and it is concluded that the final position of the tritium compound in the cells is the most important factor in the observed radiobiological effect.Notes: Zusammenfassung Die Wirkung einer kontinuierlichen Gabe von Tritium-Wasser (HTO) verschiedener Dosierung auf schwangere Ratten und ihre Nachkommenschaft wurde untersucht. Es wurde versucht, möglichst realistisch eine dauernde Strahlenbelastung während der Organogenese, wie sie auf Grund thermonuklearer Teste entsteht, nachzuahmen. Insbesondere sollte der Einfluß auf die fetale Hämopoese überprüft werden. Erhielten Ratten vom 9. bis zum 22. Tag der Schwangerschaft kontinuierlich 5800 μCi HTO/Tag, fand sich eine signifikante Erniedrigung der hämatologischen Werte bei den Neugeborenen, die während der ganzen Beobachtungszeit von 3 Wochen nach Geburt nachweisbar war. Ein Effekt auf die Wurfgröße oder Absterberate war nicht feststellbar. Niedrigere HTO-Dosen zeigten dagegen keine meßbaren Effekte auf die Hämopoese der Neugeborenen, außer einer mäßigen Lymphopenie nach Gabe von 2800 μCi/Tag. Ein Vergleich dieser Daten mit Untersuchungsergebnissen nach3H-Thymidin-Infusion ergibt, daß eine 10fach höhere Dosierung von HTO notwendig ist, um ähnliche Effekte bei der Nachkommenschaft zu ermitteln, als nach Gabe von3H-Thymidin. Die Ursache der unterschiedlich biologischen Wirkung beider radioaktiver Stoffe wird in erster Linie in der nachgewiesenen verschiedenen Position des Tritiumisotops in den Zellen gesehen. ISSN: 1432-2307Keywords: Key words Systemic amyloidosis ; Postmortem study ; Immunohistochemistry ; Classification ; Histomorphological patternSource: Springer Online Journal Archives 1860-2000Topics: MedicineNotes: Abstract Fourty-three cases of systemic amyloidosis were identified in an unselected autopsy series from our institute (6305 autopsies between 1979 and 1993) and classified immunohistochemically by means of a panel of antisera directed against five major amyloid fibril proteins. Amyloid A (AA) amyloidosis was the most common type, being found in 21 cases (48.8%). Transthyretin-derived (ATTR) amyloidosis was present in 11 cases (25.6%), and immunoglobulin light chain-derived (AL) amyloidosis in 10 cases (23.3%). A single case (2.3%) contained deposits of more than one type of systemic amyloid. AA amyoloidosis was associated with chronic inflammatory or infectious diseases (81%), malignant tumours (19%) or both (9.5%). Immunoglobulin light chain-derived amyloidoses were associated with myeloma (50%) or primary (idiopathic; 50%). In AA and AL amyloidosis the kidney was the organ most frequently involved. ATTR amyloid affecting mostly the heart and lungs presented as senile systemic amyloidosis. Systemic amyloidosis was the cause of death in 5 cases (12%) and caused symptoms in 17 cases (39%). Our results suggest that most cases can be classified by using a panel of sensitive and specific antibodies against five major amyloid fibril proteins. This technique may make amyloid type-specific therapy possible for AL amyloid patients who do not have evidence of an underlying plasma cell dyscrasia.Type of Medium: Electronic ResourceURL: -

The number of DNA synthesizing mast cells before and after regeneratory stimulus of bone marrow treated with Hydroxyurea (HU) was determined. In normal bone marrow the proliferative activity of mast cells was found very low. Animals which received 4×500 mg HU/kg at 6 hr. intervals showed a destruction of all proliferating cells. Regeneration startet 4 days after treatment and was completed 10 days afterwards. The total amount of resting cells did not change. However, the number of mast cells was increased during the first 5 days. Afterwards their number was decreased to normal levels at day 8 and later there was again an increase to about three times the level before HU administration. The latter increase between day 14 and day 16 was connected with a decrease in labelling index and labelling intensity, so that an increased DNA turnover of mast cells at that time can be concluded. The capability of mast cells to release ground substance compounds is considered to be an important factor for regenerating haemopoietic bone marrow cells.Notes: Zusammenfassung Mit Hilfe einer selektiven3H-Thymidin-Markierungsmethode (3HTdR-Markierungsmethode) zytokinetisch ruhender Zellen wurde im unbehandelten Knochenmark der Ratte und unter regeneratorischen Verhältnissen nach vorhergehender Zerstörung des Knochenmarks mit Hydroxyurea (HU) die Anzahl DNS-synthetisierender Markzellen bestimmt. Im ausgewachsenen Tier findet sich nur eine geringe Proliferationsaktivität von Mastzellen. Nach Behandlung des Knochenmarks mit 4maliger Gabe von Hydroxyurea kommt es zu einer völligen Zerstörung aller proliferierenden Zellen des Markes. Die Regeneration beginnt 4 Tage nach HU-Gabe und ist 10 Tage später vollendet. Die Gesamtzahl aller markierten Knochenmarkzellen bleibt unverändert. Innerhalb der ersten 5 Tage verdoppeln sich die Mastzellzahlen, sinken dann gegen den 8. Tag auf untere Ausgangwerte ab, um zwischen dem 14. bis 16. Tag Werte bis zum 3fachen der Zellen vor HU-Gabe zu erreichen. Auf Grund der Beobachtung eines Absinkens des Markierungsindex und der Markierungsintensität nach dem 8. bis 16. Tag kann eine gesteigerte DNS-Synthese von Knochenmarkzellen angenommen werden. Die Bedeutung der Mastzellen für die Knochenmarkregeneration wird in der Fähigkeit der Freisetzung von Metaboliten für die regenerierende Hämopoese erblickt.Type of Medium: Electronic ResourceURL: -

Before 1971 the diagnosis of abdominal involvement was based solely on physical findings and radiological studies. Treatment consisted of involved-field radiation; chemotherapy was added in selected cases. Of 15 children treated between 1963–1971 5 are still in their first remission and 2 in their second remission. Since 1971 exploratory laparotomy and splenectomy have been performed. Radiation therapy was changed from involved-to extended-field therapy and combination chemotherapy was added in all the children. No patient from this group of 16 children relapsed during a median observation time of 24 months (range 12–55 months). Such a method for the separation of these two forms of hemocyanin from Callinectes sapidus Rathbun using size-exclusion high performance liquid chromatography has been developed. The method uses columns with an exclusion limit of 1x106 daltons and monitors the binuclear copper moiety by its absorbance at 337 nm. The application of the method was demonstrated by obtaining the dodecamer to hexamer ratios for 20 hemocyanin samples from freshly caught adult intermolt male C. sapidus. The ratios ranged from 1.9:1 to 7.3:1 and appeared to be normally distributed. This variation in the polymeric composition of hemocyanin (much greater than that attributable to the reproducibility of the assay) is apparently occurring in nature, contrary to previous reports that have indicated the ratio to lie within a narrow band for a given species.Type of Medium: Electronic ResourceURL: -
