    —The distribution, localization and changes in intensity of γ-aminobutyrate transaminase (4-aminobutyrate: 2-oxoglutarate aminotransferase, EC 2.6.1.19) in rat brain have been studied during the first 20 days of postnatal life by a histochemical technique.Enzyme activity at birth was seen only in Purkinje cells of the cerebellar cortex where it increased markedly during the first 20 days. A rapid increase in enzyme activity was seen in regions of the hind-brain after 3 days but a slower increase was apparent in areas of the fore- and mid-brain. The results indicated that by 10 days post-partum nerve cell GABA-T activity had developed in the majority of brain areas studied, while glial cell GABA-T activity developed between 10 and 15 days post-partum.Evidence is presented which indicates that there is a discontinuous function of GABA-T in the developing brain.

    As a step towards indentifying exploitable differences between host and parasite at the molecular level, we have isolated and sequenced genomic clones encompassing an entire α-tubulin gene (designated α-tubulin I) from the human malaria parasite, Plasmodium falciparum. The gene, which contains two introns, encodes a product with a predicted length of 453 amino acid residues (50.3 kD). The protein sequence shows a high degree of homology to other α-tubulins, particularly that of the coccidian parasite, Toxoplasma gondii (94%), whose gene carries introns in identical positions. Only one copy of the α-tubulin I gene itself was found, although a second gene designated α-II was also identified which is closely related but which differs at both the nucleotide and amino acid sequence levels. The α-I and β-tubulin genes were found to reside on different chromosomes.

    We describe the isolation and characterization of a gene for β-tubulin from the malaria parasite, Plasmodium falciparum. This organism appears to contain a single gene encoding β-tubulin. A single transcript from this gene can be detected in the total RNA of the parasite's asexual blood stages. The complete sequence for the gene has been elucidated. It has two introns, one of which has a position identical to that of a related parasite, Toxoplasma gondii. The gene shows the usual preference for codons with A or T in the third position. The predicted amino acid sequence is compared with that of T. gondii and the human host. Further comparisons between these and fungal sequences of β-tubulins resistant to benomyl, a drug binding this protein, highlight differences that could be exploited in the development of parasite-specific anti-tubulin drugs.

    Summary The relationship between circling behavior (rotation), isolation, and aggression was investigated in normal male mice and rats. Initially the animals were tested for spontaneous nocturnal rotation, then conspecific aggression and muricidal behavior was observed for mice and rats respectively. Subsequently, animals were paired on the basis of net nocturnal rotations and either group-housed or individually housed. Four weeks later all animals were retested for the same behaviors. Spontaneous nocturnal rotation increased significantly for the isolated mice but not for the group-housed animals. Moreover, 9 of the 10 isolates became aggressive and their net rotations were significantly and positively correlated with the number of biting attacks. None of the group-housed mice became aggressive. Rats, on other hand, showed a decrease in rotation and a relationship between rotation and muricidal behavior was not evident. The possible relationship between circling behavior, aggression, and territoriality is discussed.

    Abstract We describe a girl with congenital hyperparathyroidism who presented soon after birth with respiratory distress, hypotonia, feeding difficulties, and bone deformities. Hypercalcaemia, hypophosphataemia, and raised alkaline phosphatase were present and plasma parathyroid hormone levels measured by radioassay and bioassay were raised. X-rays showed gross demineralisation with metaphyseal fractures, erosions, and sub-periostal reaction along the bones. Following surgical removal of four hyperplastic parathyroid glands and subsequent maintainance therapy with 1-alpha-hydroxycholecalciferol there has been virtually complete reversal of her bone abnormalities.
