    Continuum approximations to the discrete birth and death equations for classical nucleation are investigated. The discrete equations are parametrized by rate coefficients αi and βi for a cluster of size i to lose or gain a monomer, respectively. The continuum equations considered for the distribution function f(x,t) of clusters containing x monomers at time t are all of the form of a Fokker–Planck equation: ∂f/∂t=∂/∂x[Bf eq∂(f/f eq)/∂x], where f eq(x) is the equilibrium distribution and B(x) is a diffusion coefficient. The dependence of B(x) on various continuum approximations to the rate coefficients is discussed at length. Three different forms of B(x) are considered; that used by Frenkel [Kinetic Theory of Liquids (Oxford, Oxford, 1946)], that suggested by Goodrich [Proc. R. Soc. London Ser. A 371, 167 (1964)], and a third form proposed here. Steady state distributions and time lags obtained from the continuous and discrete equations are compared. The time-dependent Fokker–Planck equation is solved by an eigenfunction expansion and the eigenfunctions and eigenvalues of the Fokker–Planck operator are compared with those of the birth and death equations. A change of variables transforms the Fokker–Planck equation into a Schrödinger equation and permits the interpretation of the eigenvalues as energy levels in a potential function. Since the potential is approximately quadratic near its minimum, the lower eigenvalues are close to the harmonic oscillator results. The results show that the choice for B(x) suggested by Goodrich and the one proposed in this paper generally give better agreement with values from the discrete equations than the Frenkel form, used previously by most workers.

    Summary Prostate cancer is one of the most commonly diagnosed cancers and is a major cause of cancer death in men. Although the majority of the diagnosed prostate cancers will remain localized and never produce clinical symptoms during the lifetime of the host, a subset of these cancers will progress to a more malignant state requiring therapeutic intervention. Acquisition of metastatic ability by prostatic cancer cells is the most lethal aspect of prostatic cancer progression. Once this has occurred, definitive therapy is required before the initially localized metastatic cells escape from the prostate. At present, metastatic prostate cancer is incurable. Therefore, there is an urgent need to develop molecular markers that can be used to predict the metastatic potential of prostate cancers. Using somatic cell hybridization, we have demonstrated that acquisition of metastatic ability requires both the loss of metastasis-suppressor function(s) and the activation of oncogenes. In further studies using micro-cell-mediated chromosomal transfer, we located genes on human chromosome, 8, 10cen-q23, 11p11.2-13, and 17pter-q23, which, when introduced into rat prostatic cancer cells, are capable of suppressing their metastatic ability without affecting their tumorigenicity or growth rate in vivo. Initially we focused upon the human chromosome 11p11.2-13 region to clone metastasis-suppressor gene(s) positionally. One such gene, termed KAI-1, encodes a membrane glycoprotein. KAI-1 has been mapped to the p11.2 region of human chromosome 11 by fluorescence in-situ hybridization analysis. Expression of KAI-1 has been detected in all normal human tissues thus far tested, including prostate tissue. When introduced into rat metastatic prostatic cancer cells, KAI-1 significantly suppressed the metastasis without affecting the tumor growth rate. KAI-1 expression is high in human normal prostate and benign prostatic hyperplasia but is dramatically lower in cancer cell lines derived from metastatic prostate tumors.

    Summary Human tumor cells, after x-irradiation during the G2 phase of the cell cycle, show an abnormally high frequency of persistent chromatid breaks and gaps resulting from deficient DNA repair. Addition of a single human chromosome 11 from normal fibroblasts by micro-cell fusion to cell lines from six different tumors resulted in efficient repair of the radiation-induced damage to the level in normal cells. For one of the cell lines, addition of the long arm of chromosome 11 was sufficient to restore repair efficiency. In four of the six tumor lines, restoration of efficient DNA repair by chromosome 11 was associated with tumor suppression in nude mice. These results suggest that chromosome 11 carries a DNA repair gene or genes that complement the repair deficiency of tumor cells and that this gene for at least one tumor is localized to the long arm.

    A comparative study on the proton magnetic resonance of the dinucleoside monophosphates TpT, dApT, TpdA, the trinucleoside diphosphates dApTpT, TpTpdA, and poly T has been made. The purpose of this investigation was to establish whether in a trimer the proton chemical shifts of one terminal residue are influenced by the magnetic anistropy of the other terminal residue. The conformation of the trimers was first studied and shown to the similar to that of the corresponding dimer, except that of the percentage of the lefthanded conformers in the population of the trimers is probably lower than in the population of the dimers. The methylprotons, H-6, and H-1′ of the 3′-terminal T residue in TpTpdA (or the 5′-terminal T residue in dApTpT) are found to be more upfield than the same protons on the dimer TpT up to a magnitude of about 0.1 ppm. The shielding of these protons of the terminal T(s) in these trimers is even larger than those corresponding protons in the poly(T) at the same condition. From these results, it is concluded that a distant shielding effect is exerted by the 5′-terminal. A residue (or by the 3′-terminal A residue) on the other terminal residue in the trimers studied.
