    Abstract: Purified tetrameric detergent-soluble acetylcho-linesterase (DS-AChE) from human caudate nucleus was analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis in the absence as well as in presence of a reducing agent. Staining for protein revealed a main band at 66,000 daltons (light monomer) with additional bands at 78,000 daltons (heavy monomer) as well as 130,000 and 150,000 daltons (light and heavy dimers). The same four polypeptides were also detected by Western blotting and by autoradiography of [3H]diisopropylphosphoryl enzyme. Labeling of the enzyme with 3-trifluoromethyl-3-(m-[125I]-iodophenyl)diazirine showed that the heavy monomer contained the hydrophobic anchor of the enzyme, whereas the light monomer was practically not labeled. The hydrophobic anchor was susceptible to proteolytic degradation by proteinase K. The functional molarity of DS-AChE was determined by two independent methods. Four active sites for the tetrameric enzyme were estimated. The turnover number per site was 1.7 ± 107 mol of acetylthiocholine iodide hydrolyzed ± h−1.

    Pseudomorphic Si1−xGex/Si multi-quantum well structures for waveguide applications have been grown on (110) Si by molecular beam epitaxy. In a series of samples the Ge fraction x has been varied from x=0.25 to x=0.37 and x=0.50, respectively. We have used photocurrent spectroscopy on mesa diodes to demonstrate that the absorption edge of the strained Si1−xGex quantum wells can be tuned from 1.3 to 1.55 μm by the Ge fraction x. Realization and characterization of single-mode (110) Si1−xGex rib waveguides with end facets of high optical quality, prepared by cleaving, is reported. © 1995 American Institute of Physics.

    SiGe quantum wells with finite lateral size have been fabricated using local molecular beam epitaxy through shadowing masks. Scanning electron microscope and transmission electron microscope micrographs show good quality of the mesas and an effective in situ passivation by overgrowth of the buried SiGe quantum wells at the sidewalls of the mesa due to the applied growth technique. Photoluminescence measurements show clear excitonic emission from the SiGe wires and dots with lateral widths down to 2 μm. Intensities per area from the mesas are comparable to intensities from reference areas, suggesting very effective protection of the carriers against possible nonradiative recombination at the sidewalls of the mesa.

    Narrow Ge layers embedded in Si are investigated using photoluminescence (PL) spectroscopy. With increasing layer thickness a growth mode changeover from two-dimensional (2D) strained-layer growth to three dimensional Stranski–Krastanov growth is observed. Additional PL lines that are redshifted with respect to the PL signal of the 2D strained layers are attributed to islands formed by three-dimensional growth. The occurrence of these new lines is accompanied by a blueshift of the PL of the 2D layers, indicating a strong Ge diffusion from the 2D layers towards the islands. © 1995 American Institute of Physics.

    SUMMARY– The epimysial connective tissue from normal and PSE (pale, soft and exudative) porcine muscle was chemically characterized. Samples from PSE muscles exhibited markedly lower turbidity values (P 〈 0.01) than those from normal muscles. No significant differences were found in ultimate pH values. The epimysium from PSE muscles contained significantly higher (P 〈 0.05) quantities of salt soluble tropocollagen and a greater amount of heat labile collagen (P 〈 0.01) than that from normal tissues. No significant differences in α- and β-subunit composition, plasma hydroxy-proline levels or amino acid composition were evident between the two sources of epimysial connective tissues.

    Fraction I protein was extracted from fresh alfalfa (Medicago sativa) leaves and purified by ammonium sulfate fractionation, DEAE-cellulose and Sephadex G-200 chromatography. The isolate possessed a specific activity for ribulosebisphosphate carboxylase EC 4.1.1.39) of 1.01 units/mg protein. An approximate molecular weight of 573,000 was determined from values of So20,w (18.7) and Do20,w (2.97). The protein contained 26 disulfide and 37 sulfhydryl groups—only one of which was titratable in the undenatured protein complex. It migrated as a single boundry by Tiselius electrophoresis and as a dense zone in polyacrylamide gels (disc). In the presence of SDS, two principal subunits of ∼52,000 and ∼12,500 daltons were revealed. Based on the FAO/WHO (1973) provisional pattern for essential amino acids, the protein was slightly deficient in sulfur-containing residues.

    Methodology was developed that allowed sensitive measurement of phytohemagglutinin (PHA), the lectin of kidney beans. Trypsinated porcine red blood cells were treated with saline extracts of kidney beans, incubated, and the nonagglutinated cells were quantitated using a Coulter Counter. This hemagglutination activity (HA) assay was then used to monitor the PHA in cooked kidney beans. The thermal treatment (minutes @°C) required to reduce HA by 1 log cvcle was: 12 @ 100°C: 62 @ 93°C: 136 @ 88°C: 160 @ 82°C. Beans were prepared in commercially available low-temperature cookers and evaluated for tenderness and residual hemagglutinating activity.

    At least 3, and probably 4, distinct myoglobins were detected in bovine muscle by their different mobilities on starch-gel electrophoresis. The 3 major components, labeled Mb IA, Mb Ib, and Mb Ib and respectively comprising 10–15, 2–5, and 80–88% of the total myoglobin, were separated and isolated on DEAE cellulose columns employing a stepwise development technique. Various purification and separation procedures are discussed.
