    Summary Bradford andDavies did not succeed in demonstrating with their indicator-experiments that free hydrochloric acid is formed without a precursor in the oxyntic cells of the gastric mucosa. The free hydrochloric acid, as shown by them, is formed as an artefact owing to the action of the indicator.

    Abstract Different types of lattice defects have been studied by transmission electron microscopy (TEM) at 100 and 200 kV in a sample of natural marcasite. The most frequently occurring Burgers vectors are b=[001], [100] and 〈101〉. Dislocations with large Burgers vectors (b=〈110〉 and 〈111〉) have also been characterized: these are possibly split. Interactions between the different dislocations result in a build-up of dislocation networks. Dislocation structures show that the sample has undergone a strong recovery. These observations are confirmed by metallographic and scanning electron microscopy (SEM) observations after chemical etching.

    Abstract The plasma concentrations of the enantiomers of halofantrine and its N-desbutyl metabolite in six patients with malaria were measured after oral administration of 3×750 mg doses of micronised, racemic halofantrine hydrochloride given at 6-hour intervals. Significant differences were observed between the plasma concentrations of the enantiomers both of halofantrine and its N-monodesbutyl metabolite. AUC084h values were higher for (+)halofantrine (9917 μg·ml−1·h) than for (-)-halofantrine (6127 μg·ml−1·h). The clinical significance of these observations is not known. The isomers have equipotent activity in vitro but their relative toxicity has not yet been assessed.

    Summary The disposition of monodesethylamodiaquine was studied in four healthy subjects after a single oral dose of 10 mg/kg amodiaquine base. Amodiaquine was not found in any sample, but the major metabolite monodesethylamodiaquine was detected and was assumed to be the sole derivative that contributed significantly to antimalarial activity in the blood. The best fit for the decay of the metabolite was obtained with a three-compartment model. The half-lives of the first two phases were 3.2 to 11.4 h for t1/2α1 and 22.7 to 50.3 h for t1/2α2 in plasma. The half-life of the terminal phase ( t1/2β) was between 9 and 18.2 days. The concentration in whole blood was 4- to 6-times higher than in plasma. Three schedules (alternate days, weekly, daily) of the conventional prophylactic dose of 10 mg/kg per week were compared in six other healthy subjects. There were significant differences in the plasma monodesethylamodiaquine levels between the three schedules.

    Abstract The virulence of B. gingivalis strain W83 was studied in an experimental animal model. Cells grown overnight, washed and resuspended in broth, were injected intradermally or subcutaneously in the back of guinea pigs, rats and mice. This strain proved to be very virulent, causing a severe plegmonous abscess in guinea pigs. Also in mice, which are thought to be resistant to infections with black-pigmented Bacteroides strains, the same type of infection could be induced. Rats proved to be rather insensitive. The model presented can be used as a simple virulence test for these anaerobic bacteria.
