Published 2019Staff ViewPublication Date: 2019-01-15Description: In the near future, the Internet of Things is expected to penetrate all aspects of the physical world, including homes and urban spaces. In order to handle the massive amount of data that becomes collectible and to offer services on top of this data, the most convincing solution is the federation of the Internet of Things and cloud computing. Yet, the wide adoption of this promising vision, especially for application areas such as pervasive health care, assisted living, and smart cities, is hindered by severe privacy concerns of the individual users. Hence, user acceptance is a critical factor to turn this vision into reality. To address this critical factor and thus realize the cloud-based Internet of Things for a variety of different application areas, we present our comprehensive approach to privacy in this envisioned setting. Publication Date: 2018-07-27Description: In order to enhance teachers' professional awareness, it is necessary to understand and value their subjective or personal knowledge and beliefs. This study investigated the change of science teachers' personal knowledge about teaching models and modelling in science within the context of educational reform in the Netherlands. The study followed nine experienced science teachers during the first years of the implementation of a new syllabus, which emphasises models and modelling. Data collection consisted of the repeated administration of a Repertory Grid instrument. From the results, three different types of personal knowledge concerning teaching models and modelling in science were identified, each of which showed significant change over time. Type 1 combined modelling as an activity undertaken by students with the learning of specific model content. In Type 2 the learning of model content was combined with critical reflection on the role and nature of models in science. Finally, in Type 3, the learning of model content involved both students' production and revision of models, and a critical examination of the nature of scientific models in general. Publication Date: 2018-07-25Description: Kulturkonzepte sehen sich in der inter- und kulturvergleichenden Forschung zunehmend der Kritik ausgesetzt. In der Folge haben sich neue Konzepte etabliert, die auf eine Entgrenzung, Entessentialisierung, Hybridisierung und Fuzzyisierung von Kultur verweisen. Darunter befinden sich Konzepte wie das der dialektischen Perspektivenbildung, der sozialen Imaginationen (social imaginaries) und der Praxistheorie, Konzepte, die als Lösungsansätze für das immanente Theorie-Praxis Dilemma in der (inter)kulturellen Forschung und Trainingspraxis gesehen werden. Dieser Beitrag will einen Referenzrahmen für neue theoretische Ansätze bieten und zum Nachdenken über neue experimentelle Trainingsdesigns anregen.Concepts of culture have increasingly been criticized from various perspectives among the inter- and cross-cultural research communities. As a result, new conceptualisations have been developed aiming at the blurring of boundaries, de-essentialisation, hybridisation and fuzzyisation of culture. Among others, the concepts of dialectical perspectives, of social imaginaries and the theory of social practice have been proposed as some kind of solution for the immanent theory-praxis dilemma in (inter)cultural research and training. Published 2011Staff View FulltextPublication Date: 2018-07-27Description: Aim: According to the US Department of Health and Human Services (2008) and the World Health Organization (2008), regular physical activity is essential to the healthy development of children and contributes to the reduction of chronic diseases throughout their life span. However, coordinated comprehensive national and international programs to promote physical activity and sports participation are lacking. According to the German Sports Federation (2006), physical education (PE) classes at German schools are increasingly being canceled or taught outside the school. "fit for pisa" is a German intervention developed in response to the demand for scientific evaluation of interventions like daily physical education. Its goal is to provide quality management-secured, structured and standardized PE instruction by qualified instructors. Subjects and methods: The "fit for pisa" intervention has been implemented at five primary schools in Göttingen, Germany, since 2003. This daily physical education intervention is based on quality standards. At the five primary schools participating in "fit for pisa," students receive a supplementary 3 class hours of PE/week in addition to the mandatory 2 class hours/week for the entire school term. In other words, the children now receive one 45-min session of PE each school day. Daily physical education is now a part of their mandatory school curriculum. The curriculum content and teaching methods have been continuously developed and optimized based on the state guidelines for Lower Saxony. Results: Teacher training is held at the participating schools once a month to promote standardized implementation and quality management of the intervention program. Additional monthly meetings are held so that the teachers can discuss any problems, deviations or other issues occurring in the classes. Checklists are provided for the teachers and external trainers to document the content and objectives of each lesson. Supervisors evaluate the checklists at regular intervals. Using these instruments, the results of the 4-year intervention program were evaluated at the five participating primary schools. Publication Date: 2020-05-12Description: Die wirtschaftlichen und sozialen Transformationsprozesse, die in der VR China seit Mitte der achtziger Jahre zu beachtlichen strukturellen Veränderungen geführt haben, beeinflußten auch das Bildungs- und Wissenschaftssystem in einem Ausmaß, das zu Beginn der Reformperiode noch undenkbar erschien. Zu den Hauptmerkmalen, die das Bildungswesen dabei in zunehmendem Maße kennzeichneten, gehören ausgreifende Differenzierungsprozesse, begleitet von der Ausprägung neuer Hierarchien oder der weitergehenden Verfestigung bereits vorhandener. Hinzu kommt eine deutliche Regionalisierung der Ressourcenaktivierung und -verteilung, die letztlich dazu geführt hat, daß heute de facto keine verläßlichen, d.h. verallgemeinerungsfähigen Aussagen über das chinesische Bildungssystem getroffen werden können, sondern bestenfalls sind Wahrscheinlichkeiten des Antreffens bestimmter struktureller und inhaltlicher Konfigurationen zu benennen. Damit reduziert sich der Anspruch auf Realitätskonstruktion in zunehmendem Maße auf die Vorgabe von folienhaften Rahmenskizzen national relevanter Strukturen. Eine solche Folie - und das ist das erklärte Ziel dieser Arbeit - soll den Leser bei allen Einschränkungen zum Entwurf eines Bildes befähigen, das trotz seiner Unvollständigkeit maximale Annäherung an die Realität ermöglicht, ohne Anspruch auf deren umfassende Abbildung zu erheben.The economic and social transformation processes which have brought about considerable structural change in the PRC since the mid-eighties have also influenced the education and academic system to an extent which would have appeared inconceivable at the beginning of the reform period. The main features that have become more and more characteristic of the education system include extensive differentiation processes accompanied by the emergence of new hierarchies or the further consolidation of the existing ones. This is exacerbated by a distinct regionalization of the activation and distribution of resources which has ultimately led to a situation in which, de facto, no reliable, i.e. generalizable, statements can be made as to the Chinese education system as such. At best, probabilities of encountering certain structural and curricular configurations can be formulated. This reduces any attempt to convey real-life conditions to a presentation of increasingly sketchy outlines of nationally relevant structures. Two techniques for the intraoperative conservation of blood—retransfusion of the red cells using a cell-saver (CS), or retransfusion of the blood using a cardiotomy suction (CTR) system—were compared using biocompatibility markers, granulocyte activation, and production of oxygen-free radicals (OFR). In the CTR group, heparin coated circuits with an uncoated cardiotomy reservoir were used. For the CS group, identical heparin coated cardiopulmonary bypass (CPB) sets, without a cardiotomy reservoir but with a CS, were used. In each group, eight patients had coronary artery bypass grafting performed. The capacity of the whole blood and the granulocytes to produce OFR was estimated by a chemiluminescence, and granulocyte activation was measured as release of the granulocyte granule proteins myeloperoxidase (MPO) and lactoferrin. A significantly reduced capacity to produce OFR by the whole blood was noted at 45 minutes of CPB in the CTR group (68%± 17% vs 94%± 16% in the CS group). MPO release was higher after 3 hours (p = 0.05) and 20 hours (p 〈 0.05), postoperatively, in the CTR group (417 ± 77 μg/L and 257 ± 31 μg/L vs 246 ± 25 μg/L and 164 ± 12 ng/L, respectively, in the CS group). We conclude that the heparin coated CPB circuit with the uncoated cardiotomy reservoir may be less biocompatible than the identical CPB set used together with the CS.Type of Medium: Electronic ResourceURL: -

However, this phenomenon remains incompletely characterized. Using an in vitro model of neuronal/glial mesencephalic cultures, we show that the dopaminergic neurotoxin 1-methyl-4-phenylpyridinium (MPP+) stimulates the proliferation of microglial cells at concentrations that selectively reduce the survival of DA neurones. The mitogenic action of MPP+ was not the mere consequence of neuronal cell demise as the toxin produced the same effect in a model system of neuronal/glial cortical cultures, where target DA neurones are absent. Consistent with this observation, the proliferative effect of MPP+ was also detectable in neurone-free microglial/astroglial cultures. It disappeared, however, when MPP+ was added to pure microglial cell cultures suggesting that astrocytes played a key role in the mitogenic mechanism. Accordingly, the proliferation of microglial cells in response to MPP+ treatment was mimicked by granulocyte macrophage colony-stimulating factor (GM-CSF), a proinflammatory cytokine produced by astrocytes and was blocked by a neutralizing antibody to GM-CSF. Thus, we conclude that the microglial reaction observed following MPP+ exposure depends on astrocytic factors, e.g. GM-CSF, a finding that may have therapeutic implications.Type of Medium: Electronic ResourceURL: -

Recently, it was shown that S100B, a subunit of the S100 protein family, is detectable in the serum of melanoma patients and correlates with the stage of the disease in patients with metastatic melanoma. In the present study, the first evaluation of a large number of treatment observations (n = 77) in 64 different patients during chemotherapy and/or immunotherapy for advanced metastatic melanoma (stage IV) is presented. All patients received treatment according to standardized protocols comprising 8 weeks of treatment followed by routine staging procedures to evaluate therapeutic outcome. In 13 patients with tumour enlargement after first-line therapy, a second-line treatment was subsequently given. S100B immunoradiometric assay (IRMA) tests were performed before, during and after treatment at scheduled time points. In the interim analysis at 4 weeks 29 of 37 (78%) patients with tumour progression during treatment showed a raised S100B level. In the final analysis at 8 weeks, 31 of these 37 patients (84%) demonstrated rising S100B values (P 〈 0.001). Patients who responded to treatment (stable or regressing metastatic disease) showed constant or declining S100B levels in 38 of 40 patients (95%) at the interim analysis, at 8 weeks this was further increased to 39 of 40 patients (98%; P 〈 0.001). Thus, the use of S100B for monitoring treatment is adequate in the majority of cases. Our observations are of great interest for therapeutic trials of adjuvant and palliative therapies as the rise of S100B levels might indicate that re-staging and/or changes in therapy strategies should be chosen.Type of Medium: Electronic ResourceURL: -

In patients with SLE, the expression of activation markers on circulating T cells reflects disease activity. Here, we investigated whether this also holds true for patients with CLE.Patients and methods: The expression of the activation markers human leukocyte antigen (HLA)-DR and CD25 on circulating T lymphocytes was measured by flow cytometry in 24 patients suffering from different types of active CLE. Simultaneously, the disease activity was assessed clinically using a CLE activity index. Eighteen healthy donors were analyzed for control purposes.Results: HLA-DR was expressed on a significantly elevated percentage of both CD4+ and CD8+ circulating T cells in active CLE patients when compared with healthy controls. The percentage of HLA-DR-expressing T lymphocytes closely correlated with the disease activity. Interestingly, in disseminated scarring chronic discoid LE, a significantly increased percentage of CD25+ cells was observed only in the subset of skin-homing cutaneous lymphocyte antigen (CLA)+CD4+ and CD8+ T cells.Conclusion: Our results provide evidence that activation markers on peripheral blood T cells might help to objectively assess the disease activity in CLE. Furthermore, a significant population of CD25+CLA+CD8+ T cells can only be detected in a subgroup of patients with disseminated scarring CLE and might reflect the systemic expansion of activated cytotoxic T lymphocytes involved in destruction of epidermal tissue.Type of Medium: Electronic ResourceURL: -

The main focus of this paper will be current targeting concepts of melanoma (α-melanoma tumour diagnosis and internal radiotherapy) using α-MSH peptides for specific delivery of diagnostic and therapeutic radiometals. Several new α-MSH analogues (MSH1-n) were synthesized in our laboratory and conjugated to 1,4,7,10-tetraazacyclo-dodecane-1,4,7,10-tetraacetic acid (DOTA), a universal metal chelator. The resulting DOTA-MSH1-n derivatives were found to retain good binding capacity to the melanoma cell MC1 receptor (MC1R) in the low nanomolar range. In vivo tissue distribution of 5 µCi [111In]-labelled DOTA-MSH in female B6D2F1 mice with intracutaneous B16F1 melanoma tumours and with micrometastases in the lung and liver demonstrated that the radioligands accumulated specifically in the tumour tissue, reaching a maximum, for example with DOTA-MSH4, of 9.43 ± 1.06% I.D./g 4-h postinjection. Co-injection of an excess of α-MSH (50 µg) blocked the MC1Rs and hence reduced the 4-h tumour uptake by an average of 90%, which indicates that radioligand uptake by the melanoma tumours was a receptor-mediated process. Blood clearance was very rapid and 4 h after injection, the blood-associated radioactivity was as little as 0.03 ± 0.00% I.D./g. This was associated with a fast elimination of the radioactivity from all MC1R-negative tissues, except the kidneys which serve as main excretory organ. The ratios of radioactivity in melanoma tissue to that in non-target tissues 4 h after injection were all above 10 and often greater than 100, except for the kidneys. The identification of radiopeptide structures yielding reduced retention of radioactivity by the kidneys but nevertheless excellent tumour uptake is currently the main goal of our studies. The specificity of targeting melanoma metastases using radiolabelled MSH peptides was further analyzed by positron-emission tomography (PET) as well as with autoradiography of tumour tissue sections with surrounding healthy tissue after in vivo injection of the radiopeptides into tumour-bearing animals: the radioactivity was concentrated exclusively in and localized uniformly throughout the tumour tissue. Non-radioactive approaches to MC1R-mediated melanoma targeting include cytotoxic MSH–peptides, MSH–carrier conjugates and MSH–liposome constructs. These mutants displayed a multidrug phenotype and overexpressed an outer membrane protein of 50kDa, which was shown by Western blot analysis to correspond to OprN. A cosmid clone harbouring the oprN gene was isolated by partial complementation of a mutant deficient in OprM, the outer membrane component of the mexAB–oprM efflux operon. Antibiotic-accumulation studies indicated that OprN was part of an energy-dependent antibiotic-efflux system. Sequencing of a 6180bp fragment from the complementing cosmid revealed the presence of three open reading frames (ORFs), which exhibited amino acid similarity to the components of the mexAB–oprM and mexCD–oprJ efflux operons of P. aeruginosa. The ORFs were designated MexE, MexF and OprN. Mutation of the mexE gene eliminated the multidrug-resistance phenotype in an OprN-overexpressing strain, but did not affect the susceptibility profile of the wild-type strain. Expression of the mexEF–oprN operon was shown to be positively regulated by a protein encoded on a 1.5 kb DNA fragment located upstream of mexE and belonging to the LysR family of transcriptional activators. The presence of a plasmid containing this DNA fragment was sufficient to confer a multidrug phenotype onto the wild-type strain but not onto the mexE mutant. Evidence is provided to show that the mexEF–oprN operon may be involved in the excretion of intermediates for the biosynthesis of pyocyanin, a typical secondary metabolite of P. aeruginosa.Type of Medium: Electronic ResourceURL: -

Scintigraphy with the radiolabelled somatostatin analogue octreotide is a possible method for in vivo localization of the primary tumour and its metastases. Objectives To estimate the diagnostic value of indium111-octreotide scintigraphy (Octreoscan®) in detecting metastases. Methods Scans of 11 patients with Merkel cell carcinoma were evaluated, in whom scintigraphy was performed in addition to the conventional investigations, chest X-ray, ultrasonography and computed tomography (CT). Results In four cases metastases were found both by scintigraphy and by conventional methods; two investigations showed a suspicious accumulation of radioactivity on scintigraphy that could not be confirmed by CT and clinical progression. In three cases CT-verified metastases were not found by scintigraphy. Two patients were found to be tumour free, i.e. free of metastases by scintigraphy and conventional methods, indicating true-negative results. Conclusion These data confirm that scintigraphy with the radiolabelled somatostatin analogue octreotide is not clinically helpful in detecting metastases from Merkel cell carcinoma. While common among anaerobic prokaryotes, this enzyme has so far been found in only two distantly related eukaryotic lineages, anaerobic chytridiomycetes and chlorophytes. Sequence comparisons of homologues from the chytridiomycetes Piromyces and Neocallimastix, the chlorophyte Chlamydomonas, and numerous prokaryotes suggest a single, eubacterial origin of eukaryotic pyruvate formate lyases. Pyruvate formate lyase activating enzyme introduces the glycyl radical into the pyruvate formate lyase protein chain. We discovered this enzyme, which had not previously been reported from eukaryotes, in the same two eukaryotic lineages and show that it shares a similar evolutionary history to pyruvate formate lyase. Sequences with high homology to pyruvate formate lyase activating enzyme were identified in the genomes of the anaerobic protozoan parasites Trichomonas vaginalis, Entamoeba histolytica, and Giardia intestinalis. While the occurrence of pyruvate formate lyase activating enzyme together with pyruvate formate lyase in fungi and chlorophytes was to be expected, the target protein of a glycyl radical enzyme-activating enzyme in these protozoa remains to be identified.Type of Medium: Electronic ResourceURL: -

Their hydrogenosomes generate ATP from the fermentative decomposition of pyruvate but lack the hallmark pathways of mitochondrial metabolism, the TCA cycle and membrane-associated electron transport. They also, with one exception, do not contain their own genome.
