[Auszug] WITH regard to the article 'Are comets dirty snowballs or dust swarms' (News & Views 270, 558; 1978) we should like to discuss several important lines of argument and evidence that support the icy conglomerate nucleus model introduced by Whipple1 and which seem to be completely inconsistent ...

[Auszug] When leukocytes were heated to 46 C on a microscopic stage under a coverslip, pronounced pseudopods were produced, resulting in anucleated cytoplasmic fragments. The stages of this process are summarised schematically in Fig. 1. Heating resulted in the gradual polarisation of the cell into two ...

[Auszug] Venus is completely covered by a thick cloud layer, of which the upper part is composed of sulphuric acid and some unknown aerosols. The cloud tops are in fast retrograde rotation (super-rotation), but the factors responsible for this super-rotation are unknown. Here we report observations of ...

Abstract Recently we had the opportunity 10 examine and follow up over a period of 2 years an unusual case of Papillon-Lefèvre syndrome (PLS). A 10 year old boy exhibited all symptoms typical of PLS except periodontitis and premature loss of deciduous teeth. The present report aimed at studying the functional capacity of his neutiophilic granulocytes. It integrates clinical observations, histopathological findings and results of in vitro tests, The bioptic material examined included one gingival biopsy, extracted teeth, suppurative material discharged from periodontal pockets, pus emanating from a mucosal abscess, and peripheral blood leucocytes. The neutrophils were sampled on two separate occasions in two independent laboratories and tested for a variety of functions, i.e. motility, random and directional locomotion (chemotaxis), phagocytosis, membrane potential depolarization, oxygen consumption, NBT reduction, and intracellular killing of bacteria and fungi. Findings and test data indicated that in this case of PLS, neutrophilic granulocytes behaved normally with respect to all these functions including margination in blood vessels, emigration, phagocytosis of a broad range of bacteria, degranulation of lysosomes, and intracellular destruction. The data imply that factors other than neutrophil defects may be responsible for rapidly destructive periodontitis in cases where PLS is not associated with an increased susceptibility to infection.

Summary The effect of chemotactic peptides which lack chemokinetic activity has been investigated. The neutrophil response is proportional to the logarithm of the stimulus intensity, or alternatively a power function with an exponent of 0.3. Equal responses are obtained for equal ratios between the peptide concentration in the lower compartment and the threshold concentration. The significance of Weber-Fechner's law in leucocyte chemotaxis is discussed.

Abstract The short transient increase of the intracellular cAMP concentration during the first minute following stimulation, exocytosis from specific and azurophil granules, random and directional locomotion were assessed following stimulation of human and equine neutrophils with f-Met-Leu-Phe, C5ades Arg, standard gamma globulin (SGG) and the ionophore A23187. Different leucocyte-activating agents elicited distinct patterns of responses. The results showed that: (1) Chemotactic factors produced exocytosis of small amounts of vitamin B12-binding proteins but not β-glucuronidase, in the absence of cytochalasin B. (2) Chemotaxis, the appearance of the transient cAMP peak and exocytosis from specific granules in response to cytotaxins were strictly correlated in the absence of cytochalasin B but not if exocytosis was measured in the presence of cytochalasin B. Thus comparison of exocytosis measured in the presence of cytochalasin B with other functions may be misleading. (3) The non-chemotactic agents tested (SGG, A23187) produced secretion but no cAMP peak within 1 minute after stimulation, indicating that the cAMP peak is no obligatory event for triggering exocytosis in general. (4) The ionophore A23187 alone at a concentration of 10−6 M produced exocytosis from specific granules only, increased motility of cells in suspension and a marked increment of neutrophil adhesion to glass and after a lag period a sustained increase in cAMP. SGG elicited release of both vitamin B12-binding proteins and β-glucuronidase.

Summary Temporary impairment of blood supply has been suggested to cause bone remodeling. The degradation of cells and matrix and the attraction of resorbing cells were examined in this study. Bone specimens of rabbits were stored in vitro for 2–20 days. At the end of this aging process the probes were tested for their chemotactic activity toward autologous leukocytes in a diffusion chamber. Both supernatant from the aged bone specimens and ground bone particles exhibited significant chemotactic activity that was specifically attracting monocytes. It is suggested that soluble bone matrix proteins or degeneration products liberated during ischemic damage to cortical bone initiate the resorptive process.
