    Zusammenfassung Tabakprotein wurde im Ames-Test und in drei In-vivo-Tests auf Mutagenität geprüft. In den Salmonellenstämmen TA 98 und TA 100 bewirkten Methanolextrakte des Tabakproteins und der Urin von Ratten, die mit Tabakprotein gefüttert waren, erhöhte Revertantenzahlen, nicht aber Fäzesextrakte. Im Mikrokerntest wurden durchgehend schwach positive Effekte des Tabakproteins in Chinesischen Hamstern und zwei Mäuse-Inzuchtstämmen erzielt, desgleichen im Chromosomenaberrationstest und im SCE-Test an Chinesischen Hamstern. Die in den analytischen Angaben des Tabakproteins ausgewiesenen Substanzen Nikotin, Chlorogensäure und Rutin wurden einzeln im Chromosomenaberrationstest geprüft. Dabei wurde das Nikotin als der verantwortliche Faktor oder einer der verantwortlichen Faktoren für die schwach positiven Testergebnisse erkannt.

    Summary Tobacco protein was assayed for mutagenicity using the Ames test and three in vivo tests. In the Salmonella strains TA 98 and TA 100 methanolic extracts of the tobacco protein and urine of rats fed tobacco protein exhibited increased revertant numbers, but extracts of feces did not. Using the micronucleus test throughout, weak mutagenic effects after feeding the tobacco protein were detected in Chinese hamsters and two inbred strains of mice, and again in Chinese hamsters when the chromosome aberration test and the SCE test were applied. The analytical specifications of the tobacco protein listed nicotine, chlorogenic acid and rutin as components. These were examined separately in a chromosome aberration test, and nicotine was discovered to be the factor or a factor responsible for the weak positive test results.

    Abstract Exact analytical solutions are given for modal cutoff frequencies in round optical fibres with a central refractive-index depression represented by a radial index distribution Rp in the core. The cutoff frequencies Vc for modes with azimuthal order m are the solutions of Jq−1(2Vc/{2+p})=0, where q = 2m/(2 + p). An accurate analytical approximation formula for the cutoff of the LP11 mode is developed, which even circumvents the calculation of real-order Bessel function zeros. For the LP02 mode, as well as for all other circularly symmetric modes, the exact cutoff frequency is merely a linear function of the profile parameter p.

    Abstract Three substances, known to act onN-acetyl-neuraminic acid (NANA), were tested for their activity on rat peritoneal mast cells. Both neuraminidase, which splits off NANA, and the lectin fromCepaea nemoralis directed against NANA are histamine releasers. Periodate itself did not trigger the cells but inhibited the releasing action of other substances. The results provide further evidence for the role of NANA in the triggering of mast cells. A cytotoxic substance, benzyl-polyethylene imine, triggers mast cells without affecting NANA. In addition, the role of histamine-BSA conjugates as histamine releasers is described.

    Summary The authors describe transformed lymphocytes with double nuclei which occur in lymphocyte cultures after in vitro stimulation under certain culture conditions (antilymphocyte serum, washing out of the autologous plasma and haemaccel instead of serum in culture medium). In 9 tests it was possible 9 times to observe transformed lymphocytes with double nuclei. Their frequency ranged from 9 to 11% of the transformed lymphocytes with a single nucleus.

    Zusammenfassung Es werden doppelkernige transformierte Lymphozyten beschrieben, welche in Lymphozytenkulturen nach In-vitro-Stimulation unter bestimmten Züchtungsbedingungen auftreten (Antilymphozytenserum, Auswaschen des autologen Plasma und Hämaccel statt Serum im Kulturmedium). Doppelkernige transformierte Lymphozyten konnten in 9 Versuchen 9mal beobachtet werden. Ihre Häufigkeit lag zwischen 9 und 11% der transformierten einkernigen Lymphozyten.

    Abstract The protamine sulfate-induced release of histamine from rat mast cells is a non-cytotoxic reaction, similar to the 48/80-induced secretion. Polyethylene imine was found to be a less potent releaser. It is a cytotoxic substance without specificity for mast cells and acts on membranes generally. Although the two agents are related concerning their molecular weight and polybasicity, their mode of action on mast cells is clearly different.

    Zusammenfassung An insgesamt 716 Ratten und 750 Mäusen wurde eine mögliche mutagene Wirkung einer bestrahlten Nahrung geprüft. Fertilität, Tragezeitdauer, Wurfgröße, Aufzuchtleistung und Geschlechtsverhältnis zeigten in 5 aufeinanderfolgenden Rattengenerationen keine Störungen in der Reproduktionsfähigkeit an. Es konnte kein Anstieg der Mißbildungsrate bei den Rattenfeten beobachtet werden. Auch die Fruchtbarkeit der männlichen und der weiblichen Tiere der Mäusegeneration war ungestört. Durch den “dominant lethal test” an Ratten und Mäusen konnte keine embryotoxische oder mutagene Wirkung der bestrahlten Nahrung festgestellt werden, die sich in mit den verwendeten statistischen Methoden erfaßbaren Unterschieden bei den fünf untersuchten Variablen ausdrückt. Der nachgewiesene hohe Gehalt an freien Radikalen in dem bestrahlten Futter stellte nach den vorliegenden Ergebnissen keinen Faktor dar, der schädliche Wirkungen in den untersuchten Parametern bei den Versuchstieren zur Folge hatte.

    Summary The possible mutagenic effect of an irradiated diet was studied using 716 rats (5 generations) and 750 mice (1 generation). The diet consisted of 35% whole milk powder, irradiated with a dose of 4.5 Mrad, and 65% of standard rat diet. In the experiment with mice, an additional group received the same diet, but all of it was irradiated. In the rat tests the consumption of the irradiated diet did not cause any differences in fertility, duration of pregnancy, litter size, weaning index, sex ratio or number of fetal malformations. The fertility of male and female mice was also found to be unaffected. Data from 5 variables of the dominant lethal test on rats and mice were subjected to statistical evaluation and no embryotoxic or mutagenic effects were found. The high content of free radicals in the irradiated food did not give rise to harmful effects in any of the parameters studied.

    Summary Lymphocytes from 4 groups of donors were kept in Eagle's medium. The morphological change was observed on addition of purified 0-antigen (lipopolysaccharide) of S. typhi. The lymphocytes from donors with previous history of typhoid fever, from donors immunized with oral T.A.B.-vaccine and from non-immunized persons did not show any response to 0-antigen, whereas the lymphocytes from donors, vaccinated with parenteral T.A.B.-vaccine did exhibit an average transformation rate of 3 per cent.

    Zusammenfassung Die Transformationsquote in Lymphocytenkulturen von Nicht-Immunisierten, von Personen mit Typhusanamnese, sowie von parenteral und von oral Typhus-Geimpften wurde nach Stimulation mit immunologisch reinem Typhus 0-Antigen (Lipopolysaccharid) untersucht. Das 0-Antigen selbst besitzt keine Lymphocyten-stimulierende Eigenschaften. Erhöhte Transformationsquoten und Mitosezellen finden sich ausschließlich bei parenteral Geimpften, nicht dagegen bei oral Geimpften oder bei Personen mit Typhusanamnese. Im Test werden nur die durch parenterale Immunisierung sensibilisierten peripheren, ständig zirkulierenden Lymphocyten erfaßt, während nach Erkrankung oder oraler Impfung eine lokale Immunität des lymphatischen Gewebes zu diskutieren ist.
