    Abstract Nine gibberellins (GAs) have been identified from tissues of Valencia orange (Citrus sinensis Osbeck) using gas chromatography—mass spectrometry and gas chromatography-selected ion monitoring of high-performance liquid chromatography (HPLC)-fractionated extracts. These GAs are GA1, GA3, GA8, GA19, GA20, GA29, 3-epi-GA1, 2-epi-GA29, and iso-GA3. Selected-ion monitoring and stable-isotope dilution assays have been used to estimate levels of some of these GAs in vegetative and reproductive tissues. GA29 was found to be the most abundant GA measured. GA1 was found in all samples examined, and there was always less 3-epi-GA1 than GA1. GA20 was present in most extracts. Leaves of developing inflorescence shoots contained six times more GA29 than did leaves of comparable vegetative shoots. Levels of GA29 increased during the early stages of fruit development. GA20 may be more abundant in growing fruitlets than in those about to abscise; however, there were no consistent differences in the relative amounts of the other GAs. No major differences were found between tissues of immature seeded and seedless fruit, and developing seeds did not contain high levels of any of the GAs measured. It is concluded that seed-produced GAs are not essential for normal fruit development in Valencia orange.

    Summary Individual trees and stands of two provenances of Eucalyptus nitens which have marked differences in retention of juvenile foliage were studied in four plantations at different elevations. The proportion of juvenile to total foliage and growth was measured at the end of the 2nd, 3rd and 4th year from establishment. Between the ages of 2 and 4 years annual stem volume increment increased in proportion to the amount of juvenile foliage retained. By age 4 years, stem volume of trees of the juvenile persistent form was significantly larger than that of the early adult form. Increasing differences in height growth with age between provenances, which were highly significant across sites by age 4, contributed to these differences in performance. There was some evidence that the largest trees of the juvenile-persistent form were those which combined mature foliage above juvenile foliage for the longest period during the transition from juvenile to mature foliage. In the early-adult form the largest trees were those which completed the transition to mature foliage rapidly. There was no difference in the ratio of foliage mass to basal area between the two forms. It is suggested that the faster growth of the juvenile-persistent form is related to higher leaf area index and not to foliage type. A provenance of E. globulus which had a higher retention of juvenile foliage at age 4 than a second provenance had a lower stem volume, thus indicating that in this species early growth rate is not determined by foliage type.

    Abstract Potato (Solanum tuberosum L.) tuber buds normally remain dormant through the growing season until several weeks after harvest. In the cultivar Majestic, this innate dormancy persisted for 9 to 12 weeks in storage at 10° C, but only 3 to 4 weeks when the tubers were stored at 2° C. At certain stages, supplying cytokinins to tubers with innately dormant buds induced sprout growth within 2 d. The growth rate was comparable to that of buds whose innate dormancy had been lost naturally. Cytokinin-treatment did not accelerate the rates of cell division and cell expansion in buds whose innate dormancy had already broken naturally. Gibberellic acid did not induce sprout growth in buds with innate dormancy. We conclude that cytokinins may well be the primary factor in the switch from innate dormancy to the non-dormant state in potato tuber buds, but probably do not control the subsequent sprout growth.

    Abstract A radioimmunoassay, combined with high-performance liquid chromatography, has been used to analyse the zeatin-type cytokinins of potato (Solanum tuberosum L. cv. Majestic) tubers and tuber buds throughout growth and storage. During tuber growth, zeatin riboside was the predominant cytokinin detected in all tissues. Immediately after harvest, the total cytokinin concentration fell dramatically in the storage tissue, largely as a consequence of the disappearance of zeatin riboside. During storage, levels of cytokinins in the storage tissue remained relatively constant, but increased in the tuber buds. In the buds of tubers stored at 2°C there was a 20-to 50-fold increase in total cytokinin over six weeks, coinciding with the natural break of innate dormancy. At 10°C the rise in the level of bud cytokinins was slower, correlating with the longer duration of innate dormancy. Injecting unlabelled cytokinins into tubers in amounts known to induce sprouting gave rise to increases in cytokinin concentrations in the buds of the same order as the increase associated with the natural break of dormancy. Metabolism of injected cytokinins was greater in non-dormant than in dormant tubers. The roles of cytokinin concentration and the sensitivity of the buds to cytokinin in the control of dormancy are discussed.
