Published 1989Staff ViewISSN: 1365-3083Source: Blackwell Publishing Journal Backfiles 1879-2005Topics: MedicineNotes: To delineate accurately the IgG subclass distribution of thyroid auto-antibodies, sera from nine patients with Hashimoto's thyroiditis were fractionated into IgG subclasses by complete depletion of the other IgG subclasses on affinity columns. All IgG subclass fractions contained thyroglobulin and microsomal (or thyroid peroxidase) antibody activity, although when compared to the total serum concentrations of IgG subclasses. IgG4 antibodies were over-represented. However, in contrast to recent studies, this particular subclass never predominated—IgG4 antibody levels being exceeded by those of the IgG1 and IgG2 subclasses: it seems likely that these differences relate to varying sensitivity for different subclasses in previously used assay methods. This pattern of subclass activity differed from that of tetanus toxoid antibodies, which were found in six subjects. There was no light chain restriction within any subclass, showing that the overproduction of IgG4 thyroid antibodies is not of monoclonal origin. The functional affinity of subclasses for both thyroid antigens varied between patients, but IgG2 subclass fractions showed the highest functional affinity in the majority of samples. We also found that IgG2 subclass thyroid antibodies were ineffective in eliciting antibody-dependent cell-mediated cytotoxicity, as distinct from the other three subclasses. Our results show that thyroid antibodies are less restricted in their IgG subclass distribution and patients are less heterogeneous than previously described. Moreover, IgG2 thyroid antibodies are quantitatively important and differ in relative functional affinity and effector function from IgG1 and IgG4 thyroid antibodies.Type of Medium: Electronic ResourceURL: -

The antibodies were produced as mouse ascitic fluids from hybridoma cells generated from Balb/C mice immunized with purified adenovirus type 2 hexon component and crude adenovirus type 3 culture supernatants. The purified IgG fractions from all monoclonal ascitic fluids tested were identified as the IgG1 K mouse isotype. Antibody titers ranged from 102,400 to 204,800 by the IFMA, from 200 to 12,800 by indirect FA, and were generally nonreactive in counterelectrophoresis, complement fixation, hemagglutination-inhibition, serum neutralization, and immune electron microscopy titrations. The IFMA is a reliable method for quantitating low levels of specific antibody in large numbers of test samples, and is therefore ideal as a screening assay for monoclonal antibody in tissue culture fluids and in mouse ascitic fluids.Type of Medium: Electronic ResourceURL: -

Studies with HID in differing final concentrations of hydrochloric acid or sodium chloride confirmed that staining is dependent upon both the pH and the ionic strength of the dye bath and demonstrated a marked heterogeneity in the pKa of the anionic groups of sialosulphoglycoproteins. Use of the sequence High Iron Diamine—Alcian Blue pH 1.0 demonstrated that complete or almost complete staining ofO-sulphate esters occurred when HID was prepared in water (final pH 1.3). However, under these conditions HID—AB 2.5 was shown to be non-specific because only black HID staining was observed in sites containing large quantities of sialic acids. This non-specificity was due either to the masking of Alcian Blue staining by HID and/or the black HID staining of anionic groups other than sulphate. These results may account for some of the conflicting data obtained by different groups of investigators who have studied ‘transitional mucosa’ in human colonic diseases. Caution should be used in drawing conclusions from the use of HID—AB 2.5 without confirmatory evidence from other more specific procedures.Type of Medium: Electronic ResourceURL: -

The system promises free and easy movement, favourable effects on lymphoedema, and improvements with respect to dressing. Brassières are not necessarily needed. The influence of improved prosthetics on patients' self concept and well-being was investigated. A group of 67 patients after unilateral mastectomy tested custom breast forms and self-adhesive breast forms. Using the Frankfurter Selbstkonzeptionsskalen (Frankfurt Self-Concept Scales), plus additional questions concerning problems after mastectomy, self-concept was assessed before study and after 3 months. Analysis of variance with repeated measures revealed significant differences with respect to the scale Social Contact and Relations (FSKU). Most patients were better satisfied with epicutaneous self-adhesive breast forms. The weight and type of movement of the selfadhesive breast forms were considered natural, and adhesive strips were well tolerated. A total of 78% were able to wear more fashionable clothing. Breast reconstruction became less important for 62%. The new concept of self-adhesive breast forms is an improvement with respect to social and psychological rehabilitation.Type of Medium: Electronic ResourceURL: -

Lectin binding sites were characterized in the BM of gastrulating embryos of the starfish, Pisaster ochraceus. In early and mid-gastrulae, the fluorescein isothiocyanate (FITC)-lectin conjugates of concanavalin A (Con A) and wheat germ agglutinin (WGA) reveal the presence of mannose/glucose and glucosamine/sialic acid residues in the BM of all regions of the embryos. However, in the late gastrula embryo, an apparent reduction of these components is observed over the esophageal BM. Ultrastructural studies using the lectin-gold conjugates Con A, Limax flavus agglutinin (LFA), specific for sialic acid, and Dolichos biflorus agglutinin (DBA), specific for galactosamine, demonstrate that most mannose/glucose and galactosamine containing residues lie in the lamina densa, whereas most sialic acid residues are located over the lamina lucida. In addition, a statistical analysis of lectin binding in the late gastrula embryo reveals that the amount of labelling with both Con A and LFA is significantly reduced in the esophageal region, suggesting that mannose/glucose and sialic acid residuces are reduced in this region. These results confirm the observations of the FITC-lectin studies described above. They also confirm earlier studies that demonstrated a difference in BM morphology of the esophageal region (Crawford, '88). Mesenchyme cells, some of which arise from the forming coeloms (Crawford, '90), and which may represent a distinct population, colonize exclusively on this esophageal BM, where they later differentiate into muscle. ISSN: 1432-1955Source: Springer Online Journal Archives 1860-2000Topics: BiologyMedicineNotes: Abstract Investigations on the occurrence of trichodinid ciliates from fish caught in the Kiel Bight and Kiel Fjord (western Baltic Sea) were carried out between September 1996 and March 1997. Smears of the gills, fins, and skin of 120 Gadus morhua and 92 Platichthys flesus caught by fish traps and trammel nets revealed the presence of trichodinid ciliates. According to the fish species and locality, different prevalences and densities of trichodinid ciliates were found. Fish caught in the Kiel Bight revealed a lower prevalence of trichodinid ciliates on their gills (P. flesus 74.2%, G. morhua 3.8%) in comparison with fish of the same species and size caught in the Kiel Fjord (P. flesus 75.0%, G. morhua 26.2%). In both areas, P. flesus was more heavily infested than G. morhua. Seasonal changes in the prevalence of infestation of P. flesus between autumn and winter in the Kiel Fjord are proposed to be linked to an increase in bacterial biomass during winter. The fish ecology in combination with the total number of bacteria in the fish environment is discussed as an important factor influencing the abundance of trichodinid ciliates. The present data suggest the use of trichodinid ciliates as an indicator for eutrophication in brackish-water environments.Type of Medium: Electronic ResourceURL: -
