Symbiotic germination and development in vitro of Dactylorhiza majalis seeds with a strain of Rhizoctonia is very temperature dependent. Above an optimum at 23–25 °C there is a marked decline in germination percentage. Seeds that did germinate at higher temperatures had only little or no development of mycorrhiza, and developed few or no rhizoids compared with seedlings raised at optimal or lower temperatures. Six-week-old seedlings grown for additional 4 weeks on a range of temperatures had an optimal length increase at 23–24.5 °C mean temperature. At superoptimal temperatures (26 °C), the seedlings contained smaller starch reserves than those at lower temperatures and increased about as much in length as seedlings grown at 13 °C but much less than those grown at optimum. Temperature also influenced the differentiation of the leafy shoot, seedlings growing to a larger size before shoot initiation in the temperature range of optimal growth. Because of the small span between optimal and too-high temperatures, a careful assessment of temperature optimum will be necessary in any orchid/fungus relationship before judging the success of symbiosis. At optimal temperature, symbiotic germination gave a germination percentage about twice that using a good asymbiotic method. The increase in seedling length was about 45% per week in symbiotic culture compared with less than 30% in the asymbiotic culture.Type of Medium: Electronic ResourceURL: -

All the antibodies had been tested for specificity in an IUIS/WHO collaborative study, and this was confirmed by us by testing against purified myeloma proteins representing the 4 subclasses. The sera to be tested were diluted to obtain optimal sensitivity in the lower normal range for each subclass. With these serum dilutions, the lower limit of reading was 1. 2 g/l for IgG1, 0. 25 g/l for IgG2, 0.04 g/l for IgG3 and for IgG4. Age specific reference limits of the IgG subclass concentrations were determined in serum samples from 138 healthy infants and children under 14 years of age and 66 adults. The reference limits for each age group were determined by calculating the mean ± 2 SD of the logarithms to the values and then taking the antilog of the results. IgA was determined by a turbidimetric method with a reading limit of 0. 1 g/l, and the reference limits were calculated from serum samples from the 138 children under 14 years of age and from 31 healthy adults. The age specific reference limits of the IgG subclasses and IgA are given. Several infants and children had IgG4 levels below the lower reading limit. IgG subclasses were determined by a capture ELISA and IgA was quantified by turbidimetry. IgG1 concentrations were significantly lower in infants with bronchiolitis than in normal infants. The other IgG subclasses and IgA did not differ between the groups. The subgroups of infants with bronchiolitis who had previously suffered from otitis media or bronchitis, had significantly lower IgG2 than the other infants with bronchiolitis. The same was found for infants with bronchiolitis who had suffered from three or more lower respiratory tract infections. In infants who had suffered from upper or lower respiratory infections before the acute bronchiolitis, IgA was significantly higher than in infants without previous respiratory infections. Ten infants with bronchiolitis (23%) had IgGl deficiency, that is values below the lower reference limit calculated in a population of healthy Norwegian infants. No healthy infants had any IgGl deficiency. No infant with bronchiolitis had IgG2 or IgG3 deficiency. To be able to design the optimal peptides for stimulation of T cells in individuals with different HLA molecules, it is important to analyse the binding characteristics of oncogene peptides to HLA. HLA-DQ6 (DQ(α1*0102, ß1*0602)) and HLA-DRI (DR(α, ßl*0101)) molecules were purified from lysates of homozygous EBV-transformed cell lines. Purified HLA molecules were then tested for their ability to bind synthetic peptides in gel filtration assays. A p21 ras oncogene peptide (previously found to stimulate DQ6-restricted T-cell clones) and an influenza matrix peptide were labelled with 125I and served as indicator peptides for binding to DQ6 and DR1 respectively. Binding of homologous truncated and mutated p21 ras peptides and unrelated peptides was then evaluated by their capacity to inhibit binding of the indicator peptides. p21 ras-derived peptides were found to bind to both DQ6 and DR1 molecules indicating the existence of a promiscuous binding motif in these peptides. The binding affinities seemed to vary between the different peptides, but the amino acid substitutions resulting from natural mutations were not critical for binding. (Orchidaceae) was increased by illuminating surface-sterilized, rinsed and incubated seeds with white light in 16 h photoperiods. Optimal exposures (10–14 days) raised germination from 40 to 75%. Longer light treatments resulted in reduced germination percentage and smaller seedlings. Only about 2% of the seeds could actually germinate in photoperiods; the germination of the rest was delayed by initial light and required about 14 days in constant darkness. The first of these molecules confers susceptibility to celiac disease and insulin-dependent diabetes mellitus, while the two latter molecules, which share either the α chain or the nearly identical β chain with HLA-DQ(α1*0501, β1*0201), do not predispose to these disorders. The binding of peptides was detected in biochemical binding assays as inhibition of binding of radiolabeled indicator peptides to affinity-purified HLA-DQ molecules. Binding experiments with several peptides demonstrated a clear difference in peptide binding specificity between the three HLA-DQ molecules. Further, single amino acid substitution analyses indicated that the HLA-DQ molecules have different peptide binding motifs. The experimental data were corroborated by computer modelling analysis. Our data suggest that the three HLA-DQ molecules prefer large hydrophobic residues in P1 of peptides with subtle differences in side-chain preferences. HLA-DQ(α1*0501, β1*0201) and HLA-DQ(α1*0201, β1*0202) both prefer large hydrophobic residues in P9, whereas HLA-DQ(α1*0501, β1*0301) prefers much smaller residues in this position. HLA-DQ(α1*0501, β1*0201) and HLA-DQ(α1*0201, β1*0202), in contrast to HLA-DQ(α1*0501, β1*0301), prefer negatively charged residues in P4 and P7. A less prominent P6 pocket also appears to differ between the three HLA-DQ molecules. Our results indicate that polymorphic residues of both the α and the β chain determine the peptide binding specificity of HLA-DQ(α1*0501, β1*0201), but that the β chain polymorphisms appears to play the most important role. The information on peptide residues which are advantageous and deleterious for binding to these HLA-DQ molecules may make possible the prediction of characteristic features of peptide that bind to HLA-DQ(α1*0501, β1*0201) and precipitate celiac disease.Type of Medium: Electronic ResourceURL:
