The temporal relationship between the myocardial arteriovenous difference of plasma propranolol concentration and heart rate and left ventricular dp/dt maximum was examined for 30 min after intravenous administration of 10mg (0.04 mmol) dl-propranolol in nine patients with ischaemic heart disease.2. Very large positive myocardial a.v. differences of propranolol within the first 5 min became negative after 5 min, indicating a rapid, avid uptake with subsequent release of propranolol from the heart.3. The heart rate and left ventricular dp/dt maximum increased 1 min after propranolol administration and did not change after the fifth min of the observation period.4. The data suggest that a large number of unspecific binding sites for propranolol are present in the heart in addition to the specific β-receptors.

    Zusammenfassung 1. Es wurde gefunden, da\ Dialkylaminmethylole, Tetraalkylmethylendiamine und Alkyloxazolidine — Kondensationsprodukte aus Dialkylaminen und Formaldehyd — sich zur HÄrtung von Gelatine sowie zur Gerbung tierischer HÄute eignen. Reaktionsprodukte aus Formaldehyd und Monoalkylaminen bzw. Trialkylaminen besitzen dagegen diese Eigenschaft nicht. 2. WÄhrend die GelatinehÄrtung durch Formaldehyd zu einem verhÄltnismÄ\ig spröden Produkt führt, das nur eine sehr geringe KochbestÄndigkeit aufweist, erhÄlt man durch HÄrten der Gelatine mit Dialkylaminmethylolen oder Tetraalkylmethylendiaminen bzw. Alkyloxazolidinen Produkte von einer auffallend hohen KochbestÄndigkeit (bis zu 40-stündiger Kochdauer). Bei Verwendung von Alkylalkylolmethylolen bzw. Dialkylolmethylolen wird zugleich mit dem hÄrtenden Stoff eine Weichmacherkomponente an die Gelatine fixiert, wodurch der Gelatine eine mehr oder weniger hohe, graduell variierbare Geschmeidigkeit verliehen wird. Dieser Weichmacher lÄ\t sich also nicht, wie z. B. Glyzerin, aus der Gelatine herauslösen und verliert sich auch nicht im Verlaufe der Zeit. 3. Die neuen GelatinehÄrtungs- bzw. Gerbmittel, die zum Teil gleichzeitig weichmachende Wirkung besitzen, lassen sich vielseitig technisch anwenden, worüber spÄter ausführlich berichtet werden wird.

    Summary Active tubular secretion of iodamide, a water-soluble contrast medium, was demonstrated by comparing the clearances of iodamide, iothalamate and inulin. Passive tubular back diffusion was not found. The fraction excreted by tubular secretion was significantly reduced at “clinical” plasma concentrations. The glomerular filtration rate was slightly depressed at these concentrations of iodamide and diatrizoate.

    Summary The role of the sympathetic nervous system in furosemide-induced renin release was investigated in six normal subjects. After intravenous administration of furosemide, plasma renin concentrations increased more than two-fold within 15 min. Neither replacement of urinary fluid loss by intravenous infusion of saline nor pharmacological betablockade with d,1-propranolol changed the renin response to furosemide. The activity of the sympathetic nervous system, as estimated by measurement of plasma catecholamine concentrations, remained at the reference level after furosemide. It is concluded that the sympathetic nervous system is not involved in renin release after intravenous administration of furosemide.

    Summary The acute haemodynamic effects of increasing doses of parenterally administered prenalterol — a beta-adrenergic stimulating drug — were assessed in normal subjects by means of radionuclide ventriculography. Prenalterol induced dose-related increases in the left ventricular ejection fraction and the systolic pressure end-systolic volume ratio. Left ventricular end-systolic and end-diastolic volumes decreased to the same extent accounting for an unchanged stroke volume. Cardiac output increased due to a rise in the heart rate. Systolic blood pressure increased, whereas diastolic and mean blood pressure remained unchanged. Calculated total peripheral resistance decreased significantly. The maximum effect of prenalterol on cardiac performance occurred with a dose of 18 to 36 µg/kg. Plasma concentrations of prenalterol showed large interindividual variations. In conclusion, prenalterol improves the pump function of the normal heart and causes a fall in peripheral vascular resistance, implying a reduction of the load on the heart. These effects may prove beneficial in the treatment of acute heart failure.

    Summary The effect of isosorbide-dinitrate (ISDN) 20 mg 4 times daily against placebo has been tested in 12 patients with stable effortinduced angina pectoris receiving prophylactic treatment with metoprolol. ISDN did not decrease the attack rate or nitroglycerine consumption, nor was exercise tolerance increased after it. Left ventricular function, assessed by radionucleide ventriculography, increased in 6 out of 8 patients (p〈0.1). It is concluded that ISDN has no place in the treatment of haemodynamically intact patients with severe angina pectoris in spite of beta-blocking treatment, but that it may be of value in the treatment of patients with left ventricular failure, including those whose left ventricular failure has been brought about by beta-blocking treatment necessitated by angina pectoris.

    Summary The antitumor activity of titanocene dihalides (C5H5)2TiX2 with X=F, Cl, Br, I, NCS is investigated against Ehrlich ascites tumor in CF 1 mice. Varying doses of the compounds are applied as single i.p. injections 24 h pt.t. both in non-buffered (pH 1.4–3.9) and buffered (pH 4.2–5.9) solutions or suspensions, all of the substances achieving in optimum doses cure rates of 100% on day 120 p.t.t. This corresponds to I.L.S. values of 600–750% referred to the untreated controls. Some drug-induced side effects, especially the appearance of postperiotonitic symptoms several weeks after i.p. application of higher doses of titanocene dihalides, are strikingly reduced by pH elevation in the injected drug solutions.

    Zusammenfassung Die tumorhemmende Wirksamkeit von Titanocen-dihalogeniden (C5H5)2TiX2 mit X=F, Cl, Br, J, NCS wird an Ehrlich-Aszites-Tumor-tragenden CF 1-Mäusen untersucht. Verschiedene Dosen der Substanzen werden 24 h p.t.t. als einmalige i.p.-Injektionen sowohl in ungepufferten (pH 1.4–3.9) als auch in gepufferten (pH 4.2–5.9) Lösungen bzw. Suspensionen appliziert. Mit allen Verbindungen werden in optimaler Dosis Heilungsraten von 100% am 120. Tag p.t.t. erreicht. Dies entspricht Verlängerungen der mittleren Überlebensdauer (I.L.S.-Werte) um 600–750% gegenüber den unbehandelten Kontrolltieren. Einige substanzbedingte Nebenwirkungen, insbesondere das Auftreten postperitonitischer Symptome mehrere Wochen nach i.p.-Applikation höherer Dosen der Titanocen-dihalogenide, werden durch pH-Erhöhung in den Injektionslösungen deutlich zurückgedrängt.

    Summary The effect of the tumor promoter TPA and its inactive structural analogue 4-O-methyl-TPA on the induction of dorsal skin melanosis in the normal Syrian golden hamster and on the promotion of melanomas in DMBA-initiated animals was investigated. Both phenomena were observed in TPA-treated hamsters but could not be detected after exposure of animals to 4-O-methyl-TPA. In contrast to results obtained with a variety of other laboratory animals, neither TPA nor 4-O-methyl-TPA were able to induce epidermal hyperplasia of hamster dorsal skin.

    Summary A group of 150 female NMRI mice of the outbred stock Sut: NMRT was kept until they died naturally, at which time they were necropsied and examined histologically for spontaneous tumors. The natural life expectancy (median) was 782 days. Life expectancy was markedly reduced by mammary and pulmonary adenocarcinoma, and by tumors of the hypophysis. The spontaneous tumor rate was 58%. That is to say, 87 of the 150 mice had spontaneous tumors: 57 animals each had one tumor, 20 animals each had two tumors, and 10 animals each had three tumors. The organs most commonly affected by tumors were those of the lymphoreticular and haematopoietic systems, followed by the respiratory tract in second place and the breast in third place. Data reported in the literature generally show the same organ distribution, but the total tumor rate given is generally somewhat lower as the animals are seldom left alive until they die naturally (spontaneously).

Nimmt man einen ähnlichen Wirkungsmechanismus für DDP und die cancerostatisch aktiven Metallocen-dichloride TDC, VDC und MDC an, so kann der deutlich größere nicht-bindende Cl... Cl-Abstand (“bite”) von ZDC und HDC mit der cancerostatischen Unwirksamkeit der letztgenannten Verbindungen in Zusammenhang gebracht werden.

    Summary The antitumor activity of the metallocene dichlorides TDC, ZDC, and HDC against Ehrlich ascites tumor in CF 1 mice is investigated. A single i.p. injection of TDC in the optimum dose (30–60 mg/kg) 24 h p.t.t. achieves survival of 80–90% of the animals until day 180 p.t.t. without any tumor manifestation. This indicates an increase in the mean survival time of about 900% referred to the untreated animals. In contrast, ZDC and HDC exhibit no recognizable antineoplastic properties under equal experimental conditions. Assuming a similar mechanism of tumor inhibition for both DDP and the antineoplastic active metallocene dichlorides TDC, VDC, and MDC, the comparatively increased non-bonding Cl... Cl distance (“bite”) of ZDC and HDC may be responsible for the cancerostatic inactivity of the latter compounds.

    Summary The DMBA-TPA-mediated two-stage skin carcinogenesis experiment was modified in that pregnant mice were systemically treated once during pregnancy with the carcinogen. Intragastric application times were fetal days 6, 8, and 10–20. A control group of pregnant mice received repeated doses (from days 8–20) of sesame oil, which was used as a solvent for DMBA. At the age of 12 weeks, the offspring of the control group were divided into two groups, one of which was left completely untreated, the other received TPA applications over 26 weeks. The 12-week old F 1-progeny of each transplacentally initiated group was also divided into subgroups, which either received no further treatment (subgroups A) or were promoted with TPA (subgroups B). Neither the F 1-animals of the two control groups nor that of the transplacentally initiated but postnatally not promoted subgroups 6A–20A developed skin tumors. The same holds true for the TPA-promoted offsprings of mother animals which had received DMBA at days 6 and 8 of gestation. Skin tumor development after TPA promotion was first observed in animals of subgroup 10B. Thereafter, tumor rates and tumor yields increased and latency periods decreased progressively in the B-subgroups with the postponenment of initiation to later fetal periods. Day 19 of prenatal development proved to be the most sensitive period to transplacental initiation, whereas initiation at day 20 led to a significant decrease in tumor rate and yield. The capability to initiate skin tumors and the extent of initiation can be correlated to both the organogenesis of the epidermis and its proliferative rate in utero.
