    Summary The forward motility of the rat caudal epididymal spermatozoa has been studied in different Na+ concentrations. When spermatozoa were suspended in a completely Na+-free solution, the forward motility suffered a progressive fall and after 3 h was completely suppressed. This effect was fully reversible on resuspending the spermatozoa in a solution containing Na+. Amiloride caused a fall in motility and the effect was similar to that of Na+ removal. The inhibition by amiloride of the motility was concentration dependent and the dose response curve showed an IC50-value of about 5×10−5 M. The role of Na+ influx in the maintenance of sperm motility was discussed.

    Abstract  We have constructed an Aspergillus niger cDNA library with a yeast expression vector. The library DNA complemented a leucine auxotroph of Saccharomyces cerevisiae (strain BWG1-7a) at a frequency of 4×10-4. Plasmids rescued from the yeast prototrophs also complemented Escherichia coli (strain MC1066) deficient in leucine biosynthesis. Sequence determination of the rescued plasmids revealed two genes for β-isopropylmalate dehydrogenase, which we called leu2A and leu2B. Genomic-blot analysis suggested that both leu2A and leu2B were derived from single-copy genes. Northern-blot hybridization showed that in nutrient-rich medium a leu2A transcript accumulated during germination and log-phase growth while the leu2B transcript appeared late in the growth phase. In minimal medium, only leu2A expression was greatly stimulated. We examined the codon preference of these two genes. Whereas leu2A shows a bias in codon usage typical of A. niger genes, leu2B does not. These results indicate the presence in A. niger of two highly divergent, differentially regulated, isozymes for β-isopropylmalate dehydrogenase.

    Abstract It has been shown that the rat epididymis secretes proteins and organic compounds which may play a role in sperm maturation and storage. A method has been described to study caudal epididymal secretion in anaesthetized rats in vivo. The cauda epididymidis was luminally perfused with Krebs bicarbonate solution. The perfusate collected at the vas deferens was allowed to flow through a microcell and absorption at 280 nm was monitored and displayed on a recorder. Secretion as measured by this method was found to be independent of perfusion rate ranging from 2.8 to 25 μl/min and was maintained over a period of 8 h. Removal of Na+ ions from the perfusion fluid had no effect on the secretion rate. Removal of Ca2+ by perfusion of the duct with a Ca2+-free EGTA (1.5 mM) solution caused a rise in the secretion rate as revealed by an increase in absorption of the perfusate. High potassium produced similar results. Analysis of the perfusates showed that the Ca2+-free EGTA solution caused an increase in the secretion of non-protein materials whilst high potassium stimulated the release of proteins. Luminal application (0.5 mg/ml) or intraperitoneal injection (30 or 80 mg/kg) of cycloheximide had no effect on secretion during the 6 h posttreatment period. These results are discussed in relation to the possible mechanism of secretion in the epididymis.

    The type of system approximation in which the system state space x is mapped into a scalar domain V by a quadratic transformation \documentclass{article}\pagestyle{empty}\begin{document}$$ \begin{array}{*{20}c} {\rm V} & = & {\frac{1}{2}{\rm x}'{\rm Qx}} \\ \end{array} $$\end{document} where Q is appropriately determined, is used to develop a suboptimal control procedure for unconstrained lumped parameter dynamic systems via the application of Pontryagin's Maximum Principle. The optimization problem in the scalar domain becomes an initial value problem when the scalar adjoint variable is held constant through out the course of control. The resulting computational scheme includes an effective and simple way to construct the transformation matrix Q and a straightforward minimum seeking approach to locate the best constant overall average scalar adjoint parameter. For the class of problems with quadratic performance index, system equation approximation further reduces the determination of Q to the solution of a matrix Riccati equation.The application of the proposed suboptimal control procedure to four chemical engineering systems shows that the procedure is simple direct, and efficient and works particularly well for problems where the final time is large.
