    A Weissenberg camera for macromolecules with imaging plate data collection system at BL6A and BL18B stations in the Photon Factory is introduced and evaluated. The special feature of these systems is considered matching for both SR-x rays and protein crystallography. This system is user-friendly and can collect a large amount of data to higher resolution from the crystal with large unit cell dimensions. A newly developed camera can be used as a time-resolved Laue camera and a Weissenberg camera. A large image reader (IPR4080) that can scan 400×400 mm2 and 400×800 mm2 sizes of imaging plates has been developed and evaluated. The new data collection system combined with the new camera and IPR4080 will be installed at the BL18B station in October 1994. © 1995 American Institute of Physics.

    We report a neonate with angio-oedema following fetal hydrops caused by maternal parvovirus B19 infection. Levels of complement components, including total haemolytic complement activity and C1 inhibitor concentration, were within normal ranges in cord blood. Neonatal angio-oedema might be included in the clinical spectrum of parvovirus B19 infection in pregnancy.

    Design and experience of the initial operation of a new beamline (BL-16) for a 3.6-m-long, 53-pole wiggler/undulator constructed at the Photon Factory are described. The insertion device has hybrid permanent magnets with a period of 12 cm and a maximum magnetic field of 1.47 T. In the undulator mode, the energy of the fundamental peak can be varied from 40 to 400 eV. In the wiggler mode, an x-ray beam (critical energy of 6 keV) 50–100 times more intense than that from the bending magnets is obtained. The beamline consists of two time-shared branch lines: a side beamline for soft x-rays in the undulator operation, to which photon beams are deflected sideways by a cylindrical SiC mirror, and a straight line for hard x rays under the wiggler operation. On the hard x-ray line, a fixed-exit sagittal-focusing double-crystal monochromator has been installed and commissioned. Collimating and refocusing mirrors will be installed in 1989. On the undulator beamline, a soft x-ray monochromator utilizing sophisticatedly devised aberration correction optics is commissioned.

    It has been an earnest desire of protein crystallographers to collect fast, accurate, high resolution diffraction data from protein crystals, preferably with exposure time as short as possible. In order to meet this challenge, a new type of Weissenberg camera has been developed for the recording of diffraction intensity from protein crystals using synchrotron radiation. The BL6A2 line has a plane-bending mirror designed by Y. Sato. The optical bench with triangular tilt-cut Si crystal monochromator was designed by N. Kamiya and was installed in the BL6A2 hutch. The Weissenberg camera was set on the 2θ arm of the optical bench. This camera can be used with Fuji Imaging Plate (IP) as an x-ray detector, and the reading out of the image from the IP is carried out by using BA100. The characterization of this system was carried out using the native crystal of chicken gizzard G-actin DNase I complex and its Yb3+, PCMB, indium, and FMA derivatives. Since these crystals are very sensitive for x rays, the resolution limit of the diffraction was 5 A(ring) with a 4-circle diffractometer on a rotating anode x-ray generator (N. Sakabe et al., J. Biochem. 95, 887. This complex was crystallized in space group P2,2,2, with a=42.0, b=225.3, and c=77.4 A(ring). The data were collected with this system with the 430-mm radius cassette when Photon Factory was operated at 2.5 GeV and 270 mA and the wavelength λ=1.004 A(ring) was chosen. In order to avoid overlapping of diffraction spots, oscillation angle range and coupling constant (degree/mm) were settled on the basis of simulation patterns of diffraction spots up to the maximum resolution to be measured considering the direction of the crystal axes, wavelength, radius of the camera, and mosaicness of the crystal. When the oscillation axis was a axis, the oscillation angle range was selected at either 10° (1°/mm) or 5° (0.5°/mm) depending on the density of reciprocal lattice points along the incident beam, and typical exposure time in each IP was 50 and 25 s, respectively. The exposure was stopped after 10 times oscillation. The total range of 117.5° was recorded on 16 sheets of IP with an overlapping range 0.5°.The data processing was carried out using program weis coded by T. Higashi. Two complete data sets along the a and c axes were collected using two crystals, independently, and the merge R(F2) for native crystal was 0.068. In order to know the feasibility of the data (F+−F−), Patterson maps were calculated with data of each derivative, and heavy atom vectors clearly appeared as prominent peaks in the Harker sections of the Patterson maps of both Yb3+ and PCMB derivatives. The heavy atom positions were obtained from the combinations of different Patterson and different Fourier maps, and were refined by using least-squares techniques. The final figure of merit up to 2.5 A(ring) resolution was 0.61 with 22 700 reflections. The assignment of DNase I part on the electron density maps is progressing using the Nicholson molecular model referring to its structure reported by C. Oefner and D. Suck [J. Mol. Biol. 192, 605 (1986)]. In addition to promising results of ω-aminoacid pyruvate aminotransferase [N. Watanabe et al., Book of abstracts of second Japan-China Bilateral Symposium on Biophysics, p. 83 (1988)], these results support that this data collection system consisting of a new type of Weissenberg camera using SR, Fuji Imaging Plate, BA100, and program weis is one of the fastest and most accurate systems for protein crystallography in use today.     Hatchery-reared juvenile black sea bream Acanthopagrus schlegeli (Bleeker) are characterized by high lipid storage and low levels of highly unsaturated fatty acids in their bodies. In the present study, we assessed the effects of dietary fortification of the diet with eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on the improvement of physiological activity. Black sea bream 50 days old with an average weight of 0.02 g were reared for 50 days on either a control diet (commercial diet) or a commercial diet fortified with 3% EPA and DHA. The fortification with EPA and DHA reduced lipid storage in adipocytes in the intraperitoneal cavity, but decreased muscle lipid level. Consequently, the total lipid level decreased in the bodies of the fish. The proportions of EPA and DHA in muscle, liver and adipocytes were markedly increased by diet fortification, and the EPA and DHA proportions did not differ from those of wild fish. The cell diameter of adipocytes was reduced by EPA and DHA fortification. Liver function and resistance to air-dipping were improved by fortification. The results of a starvation test revealed the efficient mobilization of lipid reserves in response to energy demands prior to protein exhaustion in the EPA/DHA-enriched group. The results implied that increasing the incorporation of dietary EPA and/or DHA should be considered in the larval stage of black sea bream culture.

    Klebsiella pullulanase is a lipoprotein synthesized as a precursor with a signal peptide, which is processed by lipoprotein signal peptidase. To clarify the role of lipid modification of pullulanase, we purified lipid-modified wild-type and the unmodified (mutant) pullulanases and compared their properties. The Km and Vmax values of both pullulanases for pullulan were the same. The optimal pH and temperature, the stabilities over pH and temperature ranges, the specificity of substrates, and the patterns of inhibition of the lipid-modified and unmodified pullulanases were also the same. However, we found that the wild-type pullulanase formed trimers whereas the unmodified enzyme did not, and that the migrations of the two enzymes on sodium dodecyl sulphate/electrophoresis were different when the samples were applied on the gel without heating. The results presented in this paper and in previous work show that the correct processing and translocation of pullulanase in K. aerogenes require modification of lipid. However, the enzymatic properties and physical stabilities of pullulanase were not affected by the lipid modification.

    : The sensory evaluation of alkali-ion water and bottled mineral water was performed by panelists and their preferences for the taste of alkali-ion water was surveyed. Consequently, alkali-ion water made from tap water was found to taste better than any bottled mineral water; however, the activated carbon filtration water, which was also made from tap water, was found to taste better than alkali-ion water. Furthermore, smell, taste, feeling in the throat, aftertaste, and general impression of the mineral water, alkali-ion waters, and activated carbon filtration water were evaluated using the 1-pair comparison method. Consequently, alkali-ion water and activated carbon filtration water were more highly rated than commercial bottled mineral water.

    Background  Anti-cancer effects of cyclooxygenase (COX)-2 inhibitors have been reported, but not fully investigated in skin and oral diseases. 5-aminolaevulinic acid (ALA)-based photodynamic therapy (PDT) for treating those patients with skin and oral lesions is a highly sophisticated procedure, but the incidence of disease recurrence after treatment is rather significant.Objective  To confirm that COX-2 could be a molecular target in adjunctive therapy to ALA-based PDT, we investigated (i) COX-2 expression in various skin and oral diseases, and (ii) the inhibitory effects on cellular growth of COX-2 selective inhibitor (nimesulide), ALA-based PDT and their combination on human oral squamous cell carcinoma (SCC) cell lines.Methods  A total of 129 biopsy samples from the skin and oral mucosal lesions were tested immunohistochemically for COX-2 expression. Then the in vitro effects of nimesulide, ALA-based PDT, and their combination were determined on two SCC cell lines, HSC-2 and HSC-4. Three different methods (MTT assay, double-staining for annexin V and propidium iodide, caspase-3/CPP32 fluorometric protease assay) were applied for evaluation of their inhibitory effects on these two cell lines.Results  Among the skin diseases, a considerable number of COX-2 high expressers were found in actinic keratosis (15 of 25, 60%), Bowen's disease (13 of 17, 76%) and extramammary Paget's disease (15 of 15, 100%). In contrast, only one of 33 (3%) basal cell carcinoma tumours was a COX-2 high expresser. Among the oral mucosal biopsies, the proportion of COX-2 high expressers increased gradually from hyperplasia (one of six, 17%) through mild dysplasia (five of eight, 63%) and moderate dysplasia (20 of 23, 87%) to severe dysplasia (two of two, 100%). Nimesulide had an inhibitory effect in vitro on HSC-2 (proven to be a COX-2 high expresser), but not on HSC-4 (a COX-2 non-expresser). While ALA-based PDT showed an inhibitory effect on both HSC-2 and HSC-4, most importantly the combination of nimesulide and ALA-based PDT demonstrated a significant synergistic effect on the cellular growth inhibition of only HSC-2, but not of HSC-4.Conclusions  Our study strongly suggests that COX-2 can be one of the molecular targets in treating various skin and oral diseases. The results from our in vitro experiments also prompt us to develop a new protocol with a combination of COX-2 selective inhibitor and ALA-based PDT for more effective treatment of those diseases.
