Nineteen condylomas, 32 biopsies with slight dysplasia, 19 with moderate dysplasia and 17 with severe dysplasia-carcinoma in situ were studied. HPV status was assessed by in situ hybrization (ISH) with biotinylated probes (HPV 6/11, 16/18, 31/35/51) and the polymerase chain reaction (PCR) (HPV 16,18). DNA ploidy was measured by Feulgen DNA cytophotometry. Positivity for HPV by ISH and PCR was obtained in 48% and 59% of the biopsies, respectively. Seventy-eight per cent of the lesions were diploid or tetraploid and the remaining 22% were aneuploid. The percentage of aneuploid DNA increased with the severity of the lesions. By comparing DNA-ploidy and HPV types by ISH, diploid DNA was more frequently found in HPV 6/11 positive lesions (93%) than in HPV 16/18 positive (81%) or HPV 31/35/51 positive (57%). PCR was more sensitive for detecting HPV in aneuploid dysplastic lesions than ISH, probably indicating the HPV copies are scarce in such lesions. In summary, the results indicate some relationship between aneuploidy and HPV types 16/18 and 31/35/51, which supports an oncogenic potential of these subtypes of HPV.Type of Medium: Electronic ResourceURL: -

A 123 bp product of the 1S6110 insertion sequence specific of M. tuberculosis complex was amplified and confirmed by digestion with Sall restriction endonuclease. The time required for the procedure was 3 days. Thirty-seven samples (77.1%) were positive for M. tuberculosis complex DNA. No false positive results were obtained in nine negative controls, Of the 20 specimens tested by PCR and culture, the frequency of positivity was 90% for DNA amplification and 65% for culture. In seven cases of lupus vulgaris, the figures were 100% and 57% respectivety. In the 11 specimens culture negative or not microbiologically tesled and PCR negative, evidence for tuberculous infection was provided hy the correlation of various relative fmd absolute criteria. These results show that PCR amplification of the IS6110 insertion fragment is a rapid and accurate means for the detection of M. tuberculosis complex DNA in paraffin-embedded skin biopsies from patients with cutaneous tuberculosis, especially in paucibacillary lesions.Type of Medium: Electronic ResourceURL: -

There are two species-specific infections (fish-tank or swimming-pool granuloma, due to Mycobacterium marinum, and Buruli ulcer, caused by M. ulcerans). Most infections, however, produce a nonspecific clinical picture.Objectives To define clinical patterns of cutaneous disease in nontuberculous mycobacterial infections.Methods Fifty-one patients with cutaneous nontuberculous mycobacterial infections were reviewed. Clinical and histopathological features of normal hosts and immunosuppressed patients were compared. Two subgroups of immunosuppressed patients were distinguished: patients with cutaneous infection and patients with a disseminated infection and cutaneous involvement.Results In immunosuppressed patients the number of lesions was significantly higher. Abscesses and ulceration were also more frequently observed. Different species were found in normal hosts and immunosuppressed patients. Several clinical patterns of cutaneous infection were defined: lymphocutaneous or sporotrichoid lesions; nonlymphocutaneous lesions at the site of trauma; folliculitis and furunculosis involving the lower extremities; disseminated lesions on the extremities in immunosuppressed patients. Two patterns were observed in patients with a disseminated infection: localized cutaneous lesions and disseminated cutaneous and mucosal lesions.Conclusions Cutaneous manifestations of nontuberculous mycobacterial infections may be classified according to criteria such as cutaneous lesions and immune status.Type of Medium: Electronic ResourceURL: -

Skin biopsy of the ulcer showed granulomas, Ziehl–Neelsen stain was negative, and cultures grew Mycobacterium tuberculosis. Underlying bone and joint disease was excluded, and the lesion healed completely with 6 months of standard antituberculous treatment. In childrenMycoplasma pneumoniae and pneumococcus were identified in 79 and 29 cases respectively. Twenty-nine of 53 cases of viral infection in children were caused by respiratory syncytial virus, two-thirds of the cases occurring in children under three years of age. No children died of pneumonia. In adults pneumococcus was the most common pathogen, accounting for 81 cases. The overall mortality in adults was 7.7%. A high mortality was found in patients withHaemophilus influenzae and other gram-negative bacilli infections, and in elderly patients with pneumococcal pneumonia. Coagglutination was more sensitive than counterimmuno-electrophoresis for the detection of pneumococcal antigen in respiratory samples (p〈0.001). Counterimmunoelectrophoresis was the only useful technique for detection of pneumococcal antigen in urine specimens, concentration, overnight storage at 4 °C and specific staining significantly increasing positivity (p〈0.001).Type of Medium: Electronic ResourceURL: -

Two thousand seven hundred sixty three specimens were studied in search of an enteropathogenic bacteria, 134,1 were submited to parasitologic study and 684 to rotavirus search. Two hundred fifty one Salmonella spp., 233 C. jejuni, 152 Shigella spp., 47 Yersinia spp., 96 G. intestinalis and 181 positive Rotavirus specimens were detected. Published 1993Staff ViewISSN: 1435-4373Source: Springer Online Journal Archives 1860-2000Topics: MedicineNotes: Abstract A population-based register of cases of listeriosis admitted to acute-care hospitals has been established in Barcelona, Spain, in order to estimate the basal incidence of sporadic cases and to facilitate epidemiological surveillance of potential epidemics. Eleven acute-care hospitals reported all cases of listeriosis to a central unit following a standardized protocol. During 1990, 31 patients with listeriosis were identified, 18 of whom were residents of the city, resulting in an annual incidence of 10.95 cases per million inhabitants. Twelve of the 31 cases occurred in the period from July to September 1990, ten of them being community-acquired. The incidence of listeriosis was higher in elderly (≥ 65 years) and immunosuppressed persons. Forty-two percent of the cases were considered to be nosocomial infections. The overall mortality rate was 51.6 %. The incidence of listeriosis in the present study is one of the highest reported in the literature. A high sensitivity of the reporting system with good case identification techniques, or demographic and environmental characteristics related toListeria monocytogenes infection in our area, might be possible reasons for this geographic variation.Type of Medium: Electronic ResourceURL: -
